Resumo: This work was accomplished with the objective of studying the aspects of the infection of Colletotrichum gloeosporioides - isolated from stem and die back of coffee plants and leaves of mango; to study the development of symptoms of the pathossystem buttery x coffee plants through the evaluation of the incidence and severity of the disease; to observe the character of susceptibility between MOSPCSMM and MOSPSSMM; to confirm the relationship between C. gloeosporioides and die back in plants obtained by tissue culture and to observe in different times, to determine the isolated effects of C. gloeosporioides on the of pigments´ contents (chlorophyll the, b and total), total soluble phenols and soluble lignin in seedlings of coffee obtained by tissue culture and artificially inoculated. The experiments were carried out in DIC and in DBC. The plants were inoculated with a suspension of 2 x 106 conidia.ml-1 on the foliate area. The percentage of germination conidial and apressoria formation were evaluated at 6, 12, 18, 24, and 48 hours after the inoculation. The evaluations of the incidence and severity were accomplished at 5 days intervals, it of beginning 7 days after the inoculation with duration of 30 days. AT 3 and 7 days after inoculation were quantified contents of pigments of phenols and lignin. The spores´ germination occurred between 6 and 12 hours after the inoculation when some conidia emitted one or two germ tubes. We verified that there was significant difference to 1% of significance for the Scott-Knott test between isolated studied in this work. Besides, it showed that there was difference in the germination between isolated of MOSPSSMM. In additional there was not significant difference between isolated of MOSPCSMM studied, they presented similar effects on the medium germination of conidia. The apressoria formation was observed 6 hours after inoculation. MOSPSSMM and MOSPCSMM showed resistant to the isolated I, susceptible and moderately susceptible to isolated II. MOSPCSMM were susceptible to isolated III, while MOSPSSMM were moderately susceptible. The first necrosis symptoms in the leaves seedlings from MOSPCSMM and MOSPSSMM were only observed at seventh day after inoculation. The pathogens´ presence reduced the contents chlorophyll significantly, in the times and in the two genetic materials studied. At 7 days after inoculation of healthy and diseased plants of coffee we observed larger contents of total soluble phenols and soluble lignin in response to the attack of C. gloeosporioides.

At 7 days after inoculation of healthy and diseased plants of coffee we observed larger contents of total soluble phenols and soluble lignin in response to the attack of C. gloeosporioides.Este trabalho foi realizado com o objetivo de estudar os aspectos da infecção de Colletotrichum gloeosporioides - isolado da haste e seca de ponteiro de cafeeiro e folhas de mangueira; estudar o quadro sintomatológico do patossistema mancha manteigosa x cafeeiro através da avaliação da incidência e severidade da doença; observar o caráter de suscetibilidade entre MOSPCSMM (Mudas obtidas de sementes de plantas com sintomas da mancha manteigosa) e MOSPSSMM (Mudas obtidas de sementes de plantas sem sintomas da mancha manteigosa); confirmar a relação entre C. gloeosporioides e a seca de ponteiros e observar em diferentes tempos, determinar os efeitos de isolados de C. gloeosporioides sobre os teores de pigmentos (clorofila a, b e total), fenóis solúveis totais e lignina solúvel em mudas de café obtidas por cultura de tecidos e inoculadas artificialmente. Os experimentos foram conduzidos em DIC e em DBC. As plantas foram inoculadas com uma suspensão de 2 x 106 conídios.ml-1 sobre a área foliar. A porcentagem de germinação conidial e formação de apressórios foram estimadas a 6, 12, 18, 24, e 48 horas após a inoculação. As avaliações da incidência e severidade foram realizadas a cada 5 dias, iniciando-se aos 7 dias após a inoculação, com duração de 30 dias. Aos 3 e 7 dias após inoculação foram quantificados os teores de pigmentos, fenóis e lignina. O início da germinação ocorreu entre 6 e 12 horas após a inoculação quando alguns conídios emitiam um ou dois tubos germinativos. Entre os isolados estudados pôde-se verificar que houve diferença significativa a 1% de significância pelo teste de Scott-Knott, mostrando dessa forma que houve diferença no potencial germinativo entre os isolados em MOSPSSMM. Em relação às MOSPCSMM não houve diferença significativa entre os isolados estudados, os mesmos apresentaram efeitos semelhantes sobre a germinação média de conídios. A formação de apressórios se deu a partir das seis horas após a inoculação. MOSPSSMM e MOSPCSMM mostraram-se resistentes ao isolado I, susceptíveis e moderadamente suscetíveis ao isolado II. Em relação ao isolado III, as MOSPCSMM mostraram-se susceptíveis, enquanto que MOSPSSMM mostraram-se moderadamente susceptíveis. Os primeiros sintomas de necrose nas folhas de mudas provenientes de MOSPCSMM e MOSPSSMM, só foram observados a partir do sétimo dia após inoculação A presença do patógeno diminuiu significativamente os teores de clorofila, em relação aos tempos estudados nos dois materiais genéticos avaliados.
