Resumo: The genera Cattleya, Laelia and Brassavola occur naturally in Brazil and they are world wide wanted as ornate plants and that desire for them creates the necessity to development better propagation´s techniques not only to attend the market needs but also to contribute to the replacement of the endangered species where they could become extinct. The purpose was to test Knudson culture media concentrations, MS media vitamins, benzilaminopurine (BAP), naftalenoacetic acid (ANA), activated charcoal and banana flesh in orchid subcultivo BC Pastoral x LC Amber Glow. The explants used were from plantlets germinated in vitro with medium size 1 to 1.5 cm. The experiments were made using Knudson media concentrations (0%, 50%, 100% and 200%) combined with vitamins concentrations from MS (0%, 50%, 100% and 200%), BAP concentrations (0, 1, 2 and 4 ml L-1) combined with ANA concentrations (0.00, 0.01, 0.10 and 1.00 ml L-1) added to Knudson media; the concentrations of BAP (0, 1, 2 and 4 ml L-1) and activated charcoal concentrations (0, 200, 400 and 600 mg L-1) incorporated to Knudson media; banana cv. Nanica flesh concentrations (0, 25, 50, 75 and 100 g L-1) combined to MS media vitamins concentrations (0%, 50%, 100% and 200%). Knudson media 129% concentration enriched with vitamins from MS media 104.8% concentration promoted general plantlets growth. There is no need to add BAP and ANA to the culture media. The activated charcoal addition inhibits the leaves formation. The banana flesh 75 g L-1 concentration and activated charcoal 200 mg L-1 concentration promoted higher number of shoots formation. The banana flesh 100g L-1 concentration promoted higher medium size length on the root system and a higher plantlets fresh matter weight.
