Resumo: | In corn breeding programs factors related to the duration of the stigma receptivity, pollen grain longevity in the plant, differences in flowering periods among plants and conservation of genetic resources are aspects which reinforce the importance of researches related to factors interfering in corn pollen grains conservation. In this work, it was aimed to evaluate the conservation of corn pollen grains in different environments and water contents. The experiments were performed at the Agricultural Department of the Federal University of Lavras in the Laboratories of Seed Analysis and Molecular Biology and in an experimental field. In the pre-tests, different culture media for germination and temperatures for incubation and different times of pollen grain collection were evaluated. The tolerance of pollen grain to dissection was also determined. Following in a second experiment, pollen grains with water contents of 51.7% and 29.4% were stored in a deep freezer (-86ºC), refrigerator (4ºC) and in liquid nitrogen (-196ºC). After storage, germination and viability of the pollen grains were evaluated in vitro in a culture medium, in vivo by the staining test and through self-fecundation in plants of Le-57 line and of GNZ 2004 hybrid. The enzyme activity of pollen grains stored under the different conditions was evaluated using the enzymes esterase, malate dehydrogenase, peroxidase, superoxide dismutase and was also evaluate the expression of heat resistant proteins. Higher values of pollen grains germination were found in culture medium containing 10% of sucrose, 0.03 of Boric Acid and 0.15 of Sodium Chlorite at 25ºC and when the collection of the pollen grains was performed at 9 a.m. The conservation of the pollen grains varied with storage environment and with the water content and decreasing during storage. The germination of the pollen grains reduced with desiccation. Different patterns of enzymes and protein were observed in pollen grains under storage at different conditions. |

Different patterns of enzymes and protein were observed in pollen grains under storage at different conditions.Nos programas de melhoramento de milho, fatores relacionados à duração do tempo de receptividade do estigma, longevidade do grão de pólen na planta, diferenças no período de florescimento entre plantas e conservação dos recursos genéticos são alguns aspectos que reforçam a importância de pesquisas relacionadas aos fatores que interferem na conservação de grãos de pólen. Com este trabalho, objetivou-se avaliar a conservação de grãos de pólen de milho sob diferentes ambientes de armazenamento e teores de água. Os ensaios foram realizados no Laboratório Central de Sementes e na área experimental do Departamento de Agricultura da UFLA. Foram conduzidos ensaios preliminares, nos quais foram avaliados diferentes meios de cultura para germinação e temperaturas para incubação e diferentes horários de coleta dos grãos de pólen. Avaliou-se também a tolerância do grão de pólen à dessecação. A partir dos resultados dos ensaios preliminares, foi conduzido um experimento, no qual os grãos de pólen, com teores de água de 51,7% e 29,4%, foram armazenados em deep freezer (-86ºC), geladeira (4ºC) e em nitrogênio líquido (-196ºC). Após o armazenamento, a germinação e a viabilidade dos grãos de pólen foram avaliadas in vitro em meio de cultura, pelo teste do corante e in vivo, por meio de autofecundações em plantas da linhagem Le-57 e do híbrido GNZ 2004. Avaliou-se a atividade enzimática dos grãos de pólen armazenados nas diferentes condições por meio das enzimas esterase, malato desidrogenase, peroxidase, superóxido dismutase e proteínas resistentes ao calor. Maiores valores de germinação dos grãos de pólen foram observados em meio de cultura contendo 10% de sacarose, 0,03% de ácido bórico e 0,15% de cloreto de sódio a 25ºC e quando a coleta foi realizada às 9 horas. A conservação de grãos de pólen de milho varia com os ambientes de armazenamento e com os teores de água e torna-se reduzida com o armazenamento. O poder germinativo dos grãos de pólen reduz-se com a dessecação.
