Resumo: Jacaranda decurrens Cham. (Bignoniaceae) is a plant native to the Cerrado, popularly known as carobinha, carob or carob-of-field. Biological studies have shown giardicidal activity, antibacterial, antifungal, cytotoxic and larvicidal, being referred to the popular use in combating diarrhea, amebiasis, giardiasis, inflammations, gynecological infections, and skin disorders such as purifying the blood, indicating the presence of phenolic compounds, triterpenes and iridoid glycosides. The present study aimed to standardize extractives of leaves of Jacaranda decurrens giardicidal antioxidant potential and proven in previous studies, as parameters of quality certification. The leaves of this species were collected in São Raimundo Mangabeiras, state of Maranhão, Brazil and submitted to different extractive processes: maceration, percolation and Soxhlet with ethanol 70% (compared hydromodule 1:8, 1:10 and 1:12). Partition was held liquid / liquid leachate 1:10 hexane, dichloromethane and ethyl acetate to obtain the fractions. The extracts and fractions were subjected to phytochemical approach; levels of polyphenols were determined using the Folin Ciocalteau, total flavonoids with aluminum chloride, the antioxidant activity was performed by DPPH and evaluation giardicidal activity "in vitro" cytotoxicity by colorimetric method direct and indirect emoregando axenic strains of Giardia lamblia strain Portland-1 (ATCC30888). The profiles of the extractives were obtained by spectrometry UV / Vis, HPLC / UV / Vis and LC / MS. The results showed the presence of phenolic substances, and lack of triterpenoid steroids and alkaloids. The content of total polyphenols in the extracts and fractions ranged from 13.10 to 80.41% and flavonoids, 1.74 to 5.15%. The extracts and fractions showed antioxidant activities between EC50 ranges from 26.39 to 339.73 mg / mL. Extracts (M110, P108 and S112) were assessed giardicidal activity showed inhibitory activity against trophozoites of Giardia lamblia, and the values found between 17.10 to 18.92 ug / ml. The analysis of UV spectra showed similarities with maximum absorptions between 230 nm to 300 nm, indicating the presence of flavonoids. Analyzing the chromatograms, attributed to the percolation method in respect of 1:10 for the best extraction Jacaranda decurrens. Chromatogram obtained by HPLC / UV / Vis at a wavelength of 254 nm, it was found that the hexane fraction there was a peak coincident with the pattern of oleanolic acid with the same retention time (108.403), suggesting that this acid is one the chemical constituents of the plant. With mass spectrometry, we identified rutin, quercetin-pentosídeo-hexosídeo, quercetin-3-β-glucoside, arjunolic acid and acid-2α hidroxioleanólico. We conclude that the triterpenes (oleanolic acid) and flavonoids are likely responsible for giardicidal and antioxidant action.

(Bignoniaceae) é uma planta endêmica do Cerrado, conhecida como carobinha, caroba ou caroba-do-campo. A espécie é empregada popularmente no tratamento de amebíase, giardíase, inflamações, infecções ginecológicas, afecções cutâneas, no combate a diarreia e como depurativa do sangue, sendo comprovadas as ações giardicida, bactericida, antifúngica, citotóxica e larvicida, com a presença de compostos fenólicos, triterpenos e iridoides glicosilados. O presente trabalho teve como objetivo padronizar extrativos de folhas de Jacaranda decurrens com potencial giardicida e antioxidante, como parâmetros de certificação de qualidade. As folhas da espécie foram coletadas no município de São Raimundo das Mangabeiras, estado do Maranhão, Brasil e submetidas a diferentes processos extrativos: maceração, percolação e Soxhlet com etanol 70% (relação hidromódulo 1:8, 1:10 e 1:12). Foi realizada partição líquido/líquido do percolado 1:10 em hexano, diclorometano e acetato de etila para a obtenção das frações. Os extratos e frações foram submetidos à abordagem fitoquímica; os teores de polifenois foram determinados com reagente de Folin Ciocalteau; os flavonoides totais com cloreto de alumínio; a atividade antioxidante foi realizada pelo método DPPH e a avaliação da atividade giardicida ―in vitro‖ de citotoxicidade pelo método colorimétrico direto e indireto, emoregando cepas axênicas de Giardia lamblia, linhagem Portland-1 (ATCC30888). Os perfis dos extrativos foram obtidos por espectrometria de UV/Vis, CLAE/UV/Vis e LC/MS. Os resultados demonstraram presença de substâncias fenólicas, triterpênicas e ausência de cumarinas, esteroides e alcaloides. Os teores de polifenóis totais nos extratos e frações variaram de 13,10 a 80,41% e os de flavonoides, 1,74 a 5,15%. Os extratos e frações apresentaram atividades antioxidantes entre os intervalos de CE50 26,39 a 339,73 μg/mL. Os extratos (M110, P108 e S112) submetidos à avaliação da atividade giardicida demonstraram atividade inibitória sobre os trofozoítos de Giardia lamblia, sendo os valores encontrados entre 17,10 a 18,92 μg/ml. As análises dos espectros de UV demonstraram semelhanças com absorções máximas entre 230 a 300 nm, indicando a presença de flavonoides. Analisando os cromatogramas, atribuiu-se ao método de Percolação na relação de 1:10 o melhor extrativo para Jacaranda decurrens. Pelo cromatograma obtido por CLAE/UV/Vis no comprimento de onda de 254 nm, identificou-se que na fração hexânica houve um pico coincidente com o padrão do ácido oleanólico com o mesmo tempo de retenção (108.403), sugerindo que esse ácido seja um dos constituintes químicos da planta. Com o espectrômetro de massas, identificou-se ácido cafeoil, rutina,quercetina-pentosídeo-hexosídeo, quercetina-3-β-glicosídeo, ácido arjunólico e ácido 2α-hidroxioleanólico. The extracts and fractions showed antioxidant activities between EC50 ranges from 26.39 to 339.73 mg / mL. Extracts (M110, P108 and S112) were assessed giardicidal activity showed inhibitory activity against trophozoites of Giardia lamblia, and the values found between 17.10 to 18.92 ug / ml. The analysis of UV spectra showed similarities with maximum absorptions between 230 nm to 300 nm, indicating the presence of flavonoids. Analyzing the chromatograms, attributed to the percolation method in respect of 1:10 for the best extraction Jacaranda decurrens. Chromatogram obtained by HPLC / UV / Vis at a wavelength of 254 nm, it was found that the hexane fraction there was a peak coincident with the pattern of oleanolic acid with the same retention time (108.403), suggesting that this acid is one the chemical constituents of the plant. With mass spectrometry, we identified rutin, quercetin-pentosídeo-hexosídeo, quercetin-3-β-glucoside, arjunolic acid and acid-2α hidroxioleanólico. We conclude that the triterpenes (oleanolic acid) and flavonoids are likely responsible for giardicidal and antioxidant action.
