Resumo: | In developing countries traditional medicine based on the use of plant extracts play a majorrole in health care. Arrabidaea chica (HBK) Verlot (Bignoniacea) is a common plant in theAmazon region. Its leaves are widely used in folk medicine for wound healing, treatment ofinflammation, intestinal colic, blood dysfunction, anemia, hemorrhage and leukemia. The antiinflammatoryeffect of its extract was already demonstrated in vitro and in vivo. The aim ofthe present study was to evaluate the cytotoxic activity of extracts of A. chica against humantumor cell lines. Also evaluate the in vivo antitumor activity in mice bearing solid Ehrlichtumor by hematological and biochemical studies, evaluation of the development of the tumorand immunophenotyping of mononuclear inflammatory cells in blood and associated withtumor tissue. The anti-inflammatory activity was evaluated using the murine sponge model.Methanol, ethanol and aqueous extracts were obtained from dried lives of A. chica. Thepresence of the flavonoid kaempferol was verified by high performance liquidchromatography. The ethanol extract (EE) showed cytotoxic activity against Jurkat and HL60leukemic cells, with IC50 values of 27.9 mg / mL and 26.9 mg / mL, respectively. Also,ethanol extract-EE induced DNA fragmentation assessed by flow cytometry assay, suggestingthat the toxicity presented may be related to induction of apoptosis. The ethanol-EE and CA3extracts, administered orally at a dose of 300mg/kg presented anti-inflammatory activityassessed by the murine sponge model. Both extracts inhibited neovascularization andrecruitment of neutrophils to the sponges without altering the macrofage mobilization andvascular endothelial growth factor (VEGF) concentration. There was a reduction in solidtumor volume in mice treated with EE and CA3 (30mg/kg) after 10 days of daily oraladministration (p<0,05) without changes in hematological and biochemical parameters. Theethanol extract-EE increased neutrophils and 1 and globulins, and decreased 2 globulins.The ethanol-EE and CA3 extracts reduced the percentage of T CD4+ cells on blood withoutaltering TCD3+, NK and B lymphocytes. The aqueous fraction CA3 showedimmunomodulatory activity by reducing the number of TCD3+, CD8+ and NK cells in tumortissue, without altering the percentage of B and T CD4+ cells. The extracts did not producetoxic effects in mice in either study. It is concluded that the ethanol extract and aqueousfraction CA3 of A. chica have antitumor effect after oral administration, confirming its use intraditional medicine. The activity of the ethanol extract-EE seems to be related to the potentcytotoxic and pro-apoptotic activities, as well as its anti-inflammatory and antiangiogenicactivities. The antitumor effect presented by the aqueous fraction CA3 is possibly related totheir anti-inflammatory and antiangiogenic activities. These results suggest biotechnologicalpotential of the extracts of A. chica for obtaining new antitumor drugs. |

AArrabidaea chica (HBK) Verlot (Bignoniacea) é uma planta comum na região amazônica esuas folhas são utilizadas na medicina popular para cicatrização de feridas, tratamento deinflamações, cólicas intestinais, hemorragia, anemia e leucemia. Seu efeito antiinflamatório jáfoi demonstrado in vitro e in vivo, entretanto não foram encontrados relatos na literaturaquanto à sua atividade antitumoral. O objetivo do presente estudo foi avaliar a atividadecitotóxica dos extratos etanólico, metanólico e aquosos da A. chica em linhagens de célulastumorais humanas e em camundongos Swiss portadores do carcinoma sólido de Ehrlich.Foram preparados diferentes extratos a partir de folhas secas da A. chica extraídos com água,metanol ou etanol e verificada a presença de flavonóides pela cromatografia líquida de altaeficiência. Em estudos in vitro, o extrato etanólico (EE) apresentou atividade citotóxica contraas células tumorais Jurkat, HL60 e MCF-7, com valores de IC50 para Jurkat e HL60 de27,9g/mL e 26,9g/mL, respectivamente. Ensaios de citometria de fluxo demonstraram queesse extrato (EE) induziu fragmentação do DNA celular, sugerindo que a toxicidadeapresentada pelos compostos pode estar relacionada com a indução de morte por apoptose.Nos estudos in vivo, conduzidos em camundongos portadores do tumor sólido de Ehrlich,foram realizados ensaios bioquímicos e hematológicos, avaliação do desenvolvimento tumorale imunofenotipagem por citometria de fluxo das células inflamatórias mononuclearespresentes no sangue e tecido tumoral. Os extratos EE e CA3, após 10 dias de administração oralna concentração de 30mg/kg, causaram redução no volume do tumor sólido de Ehrlich(p<0,05), sem causar alterações nos parâmetros hematológicos e bioquímicos plasmáticos. Ogrupo tratado com o extrato EE apresentou aumento dos neutrófilos e das 1 e globulinas, ediminuição das 2 globulinas. A imunofenotipagem das células mononucleares no sangue,demonstraram que os extratos EE e CA3 da A. chica reduzem a porcentagem de linfócitos TCD4+sem alterar o número total de linfócitos TCD3+, NK e linfócitos B. O extrato CA3 apresentouatividade imunomoduladora diminuindo o número de células TCD3+, TCD8+ e células NK notecido tumoral, sem alterar o percentual de linfócitos B e TCD4+. Ainda neste contexto,também foi possível avaliar a atividade antiinflamatória dos extratos etanólico-EE e CA3 emmodelo murino de implante de esponja. Os extratos EE e CA3, administrados por via oral, nadose de 300mg/kg inibiram a neovascularização e o recrutamento de neutrófilos para asesponjas implantadas. Os extratos não produziram efeitos tóxicos nos camundongos nas dosesutilizadas em nenhum dos estudos. Conclui-se que o extrato etanólico e a fração aquosa CA3 da17Arrabidaea chica apresentam efeito antitumoral após a administração oral, confirmando o seuvalor terapêutico como indicado pela medicina popular. A atividade do extrato EE parece estarrelacionada ao potencial citotóxico e pró-apoptótico, bem como seu potencial antiinflamatórioe antiangiogênico, confirmados nos estudos in vivo no modelo de Ehrlich. A ação antitumoralda fração aquosa CA3 está possivelmente relacionada à sua atividade antiinflamatória eantiangiogênica.
