Resumo: | Temperature is one the most important abiotic factors for the development of biologic cicle of entomopathogeni fungus. The pathogenic fungus has developed molecular mechanisms like protein expression related to growth at high temperature to survive at body temperature of the host. Thus, the aim of this study is to analyse the effect of temperature on in vitro growth of the C. lamprauges and to compare the pathogen cultivated at two different temperatures (28oC and 37oC) by proteomic analysis. Five isolated of C. lamprauges from sick sheep were used to analyse the growth at different temperatures. It was inoculated spore suspension at petri dish with 2% Dextrose Sabouraud Agar, they were incubated at 20, 25, 30, 35 e 40oC and the radial growth was measured daily. For the proteomic analysis, C. lamprauges was cultivated at 28oC and 37oC, it was performed the protein extraction and the two- dimensional polyacrylamide gel. The experiment showed that the isolated grown to approximately 35oC and before, stop the growth. There was inhibition of growth at 18 and 42°C. After differential proteome analysis, a total of 51 and 49 spots were found at 28 and 37oC, respectively, 16 spots were differentially expressed, seven (7/16) were down expressed and nine (9/16) were over expressed at 37oC compared with 28oC. |

Para sobreviver na temperatura corpórea de seu hospedeiro, os fungos patogênicos têm desenvolvido mecanismos moleculares importantes, como a expressão de proteínas relacionadas ao crescimento em altas temperaturas. Assim, o objetivo desse trabalho foi analisar o crescimento in vitro do C. lamprauges em diferentes temperaturas e comparar, através da análise proteômica, o patógeno cultivado em duas temperaturas distintas, sendo uma considerada baixa (28oC) e outra alta (37oC) em relação à temperatura ótima de crescimento obtida nesse experimento. Para análise do crescimento em diferentes temperaturas, foram utilizados cinco isolados de C. lamprauges, oriundos de ovinos doentes. Após inoculação da suspensão de esporos em placas de Petri contendo Agar Sabouraud dextrose 2%, estas foram incubadas a 20, 25, 30, 35 e 40oC e o crescimento radial foi medido a cada 24 horas. Para análise proteômica, o fungo foi cultivado a 28oC e a 37oC, realizou-se a extração de proteínas e confecção de géis de poliacrilamida bidimensionais. Observou-se que os isolados cresceram até uma temperatura de aproximadamente 35°C e após, reduziram seu crescimento. Houve inibição do crescimento a 18 e 42oC.
