Resumo: | Pilosocereus gounellei popularly known as xique-xique traditionally been used by the northeast population for the treatment of prostatic inflammation. Its wide use in folk medicine justifies the toxicity studies to ensure the safety of its use. In this sense we propose to conduct a pre-clinical toxicological evaluation of the ethanol extract of Pilosocereus gounellei K. Schum (Pg-EEtOH) following the guide for the conduct of non-clinical toxicology and safety pharmacology necessary for the development of drugs (01/2013). For this bioassay was performed using Artemia salina to determine the LC50 and were also Wistar rats and Swiss mice of both sexes for toxicological analysis in order to verify the effects of the extract on the physiological parameters of these animals. In the acute toxicology test is used a single dose of 2000 mg / kg orally after treatment are observed for 14 days for the following parameters: the behavioral effects at 72 hours; consumption of water and food daily; as well as the weekly weight gain. After the 14th day will determine the LD50. The surviving animals will be sacrificed for analysis of biochemical, hematological, macroscopic evaluation of organs and histopathology in altered organs. In the chronic toxicity study will be administered divided doses of Pg-EEtOH 90 days will be assessed the following parameters: temperature, blood sugar, consumption of water and food, weight gain, exploratory and motor activity of animals, hematological and biochemical parameters of rats in study. The genotoxicity was evaluated by the micronucleus test in peripheral haematological cells in vivo Rodents. The antiinflammatory activity was assessed by paw edema model induced by carrageenin. Pg-EEtOH showed a LC50 of 547.3 g / ml, an LD50> 2000 mg / kg did not cause behavioral changes, caused a significant reduction in the consumption of water and feed. In females it was observed an increase in serum levels of urea and a decrease in creatinine levels. The Pg-EEtOH did not change the temperature or blood glucose of significantly animals also did not change the consumption of water and food and not the weight gain. Behavioral analysis of animals no significant changes were not found in the evaluated parameters also there has been no change in the autonomic motor system of these animals. The biochemical and hematological parameters there were no significant change when compared to the control group. In micronuclei research we can see that the extract did not induce micronuclei formation compared to the control group. In the paw edema model it was observed that only the dose of 25 mg / kg significantly reduced the edema. Based on these data we observe that the Pg-EEtOH has a low toxicity, and genotoxicity besides having anti-inflammatory activity. |

Neste sentido nos propomos realizar uma avaliação toxicológica não-clínica do extrato etanólico de Pilosocereus gounellei (fac weber) (Pg-EEtOH) seguindo o guia para a condução de estudos não clínicos de toxicologia e segurança farmacológica necessários ao desenvolvimento de medicamentos (01/2013). Para tanto foi realizado o ensaio biológico utilizando Artemia salina para determinar a CL50 e também foram utilizados ratos Wistar e camundongos Swiss de ambos os sexos. Inicialmente foi realizada uma triagem farmacológica com intuito de verificar os efeitos do extrato sobre os parâmetros fisiológicos desses animais. No ensaio toxicológico agudo foi utilizada a dose única de 2000 mg/kg por via oral, após o tratamento foram observados durante 14 dias os seguintes parâmetros: os efeitos comportamentais durante 72 horas; consumo de água e ração diariamente; bem como a evolução ponderal semanalmente. Após o 14 dia foi determinada a DL50. Os animais sobreviventes foram sacrificados para análise dos parâmetros bioquímicos, hematológicos, avaliação macroscópica dos órgãos e exames histopatológicos nos órgãos alterados. No estudo toxicológico crônico foram administradas doses fracionadas do Pg-EEtOH durante 90 dias, foram avaliados os seguintes parâmetros: temperatura, glicemia, consumo de água e ração, evolução ponderal, atividade exploratória e motora dos animais, parâmetros hematológicos e bioquímicos dos ratos em estudo. A genotoxicidade foi avaliada através do teste do micronúcleo em células hematológicas periféricas de roedores in vivo. A atividade antiinflamatória foi avaliada através do modelo de edema de pata induzida por carragenina. O Pg-EEtOH apresentou uma CL50 de 547,3 µg/mL, uma DL50 > 2000 mg/kg, não promoveu alterações comportamentais, porém induziu uma diminuição significativa no consumo de água e ração. Nas fêmeas foi observado um aumento nos níveis de séricos de ureia e uma diminuição nos níveis de creatinina. No ensaio crônico o Pg-EEtOH não alterou de forma significativa a temperatura, a glicemia, a evolução ponderal, o consumo de água e ração dos animais. Na analise comportamental não foi observado alteração significativa nos parâmetros avaliados também não ocorreu nenhuma alteração no sistema motor autônomo desses animais. A analise dos Parâmetros bioquímicos e hematológicos não apresentou alteração significativa quando comparado ao grupo controle. Na pesquisa de micronúcleos pode-se observar que o extrato não induziu a formação de micronúcleos quando comparado com o grupo controle. No modelo de edema de pata foi observado que apenas a dose de 25 mg/kg reduziu significativamente o edema.
