Resumo: | The search for better conditions to preserve biomaterials to transplantation has received attention in the last years, and the efficiency of trehalose has been proven in many experiments, due to its particular capability to stabilize living organisms, frequently under environmental stress. Several studies in theoretical chemistry has been performed, trying to elucidate such protection mechanism, when particularly related to dehydration. However, the theoretical descriptions used seem not to be suitable to these systems, where eletrostatic interactions are determining. Therefore, in this work, a phosphocholine molecule is studied, since it can be considered a very good prototype of the hydrophilic portion os the phospholipids membrane. The hydrophilic portion is the location where the interaction trehalose-membrane is supposed to happen. For systems like that, where a molecular charge separation is present, electronic correlation effects have to be considered. B3LYP/6-31G(d,p) calculations were performed in a conformational study where dihedral angles ?1, ?2, ?3, ?4 and ?5 of phosphocholine were scanned in the gas-phase and aqueous solution. Conformational maps were obtained for the isolated and solvated system, and the most stable conformations found from these maps. |

Sendo assim, existe uma grande demanda por compostos capazes de conservar ?rg?os e tecidos por um maior per?odo de tempo. Neste contexto, tem-se avaliado a utiliza??o do dissacar?dio trealose como agente protetor nestes processos, dada a sua conhecida capacidade de estabilizar materiais biol?gicos em diferentes condi??es de estresse. Diversos estudos de qu?mica te?rica t?m sido feitos a fim de tentar elucidar poss?veis mecanismos atrav?s dos quais a trealose confere prote??o a materiais biol?gicos durante per?odos de estresse, com particular interesse no processo de desidrata??o. No entanto, as descri??es te?ricas utilizadas parecem n?o ser muito adequadas para o tratamento de sistemas deste tipo, onde intera??es de natureza eletrost?tica parecem ser respons?veis pela intera??o trealose-membrana celular. Sendo assim, neste trabalho, prop?e-se um prot?tipo de membrana fosfolip?dica que representa a por??o hidrof?lica de fosfolip?dios de membrana potencialmente capaz de descrever as superf?cies celulares, uma vez que acredita-se que as intera??es trealose-membrana se estabele?am, basicamente, entre este dissacar?dio e as por??es hidrof?licas dos fosfolip?dios. Sabendo que para sistemas como este, com forte separa??o de cargas, os efeitos de correla??o eletr?nica n?o podem ser negligenciados, utilizou-se um m?todo quanto-mec?nico, baseado na Teoria do Funcional da Densidade, para a descri??o desta por??o de fosfolip?dio. Atrav?s de c?lculos B3LYP/6-31G(d,p), realizou-se o estudo conformacional dos ?ngulos ?1, ?2, ?3, ?4 e ?5 da metilfosfocolina, tanto em fase gasosa como em solu??o.
