Resumo: This study investigated methods of diagnosis and control of Fasciola hepatica in ruminants, in addition provid information about the population dynamics of Lymnaea columella. In the first stage of labor, between November 2010 and June 2013, the mollusc species L. columella were collected at five diferent locations and comper with Meteorological data such as precipitation and temperature. There was no significant diference between the mean number of mollusks at the seasons. The second stage of the study involved the comparison of the sedimentation technique described by Foreyt (2005) with four sieves technique proposed by Gir?o; Ueno (1985) with artificially infected cattle feces with F. hepatica eggs for quantitative diagnosis of this parasite. After statistical analysis, the sedimentation technique showed better results with higher percentages of recovery in addition to having lower cost and be easier to perform than the sieving technique. In the third and final stage of this study, adult specimens of F. hepatica were collected from sheep previously treated with albendazole and one sheep control. The evaluation times were 48, 72 and 96 hours post- treatment. Flukes were processed for histopathologic and assessed for changes in male reproductive sistem. It was observed that during the evaluation period were significant changes in spermatogenic cells which include rarefaction cell, apoptosis in cells with typical pyknosis and karyorrhexis and thereby decrease sperm production, as well as intra-and extracellular vacuolation. These results demonstrate that albendazole altered sperm production which interferes with the production of fluke eggs, demonstrating the effectiveness of the drug in this experiment. Studies of seasonality intermediate host of F. hepatica, which contribute to the choice of the best diagnostic technique of this parasite and demonstrating efficacy and / or drug resistance are essential for the establishment, in each region, an efficient strategic program of fluke control.

Na primeira etapa do trabalho, entre novembro de 2010 e junho de 2013, moluscos da esp?cie L. columella foram coletados em cinco diferentes pontos e correlacionados a dados meteorol?gicos como precipita??o e temperatura. N?o houve diferen?a entre o n?mero m?dio de moluscos nas esta??es do ano. A segunda etapa do trabalho envolveu a compara??o da t?cnica de sedimenta??o descrita por Foreyt (2005) com t?cnica de quatro tamises proposta por Gir?o; Ueno (1985) com infec??o artificial de fezes de bovinos com ovos de F. hepatica, para diagn?stico quantitativo deste parasito. Ap?s an?lise estat?stica a t?cnica de sedimenta??o apresentou resultado superior com maiores percentuais de recupera??o al?m de possui menor custo e ser de mais f?cil execu??o que a t?cnica de tamisa??o. Na terceira e ?ltima etapa deste trabalho, exemplares adultos de F. hepatica foram coletados de ovinos previamente tratados com albendazole e de um ovino controle. Os tempos de avalia??o foram 48, 72 e 96 horas p?s-tratamento. Os parasitos foram processados por meio de t?cnica histopatol?gica e avaliados quanto ? altera??es no aparelho reprodutor masculino. Observou-se que no decorrer do tempo de avalia??o houve altera??es significativas nas c?lulas espermatog?nicas as quais incluem: rarefa??o celular, apoptose com c?lulas apresentando caracter?sticas de picnose e cariorrexe e, consequentemente, diminui??o da produ??o de espermatozoides, al?m de vacuoliza??o intra e extracelular. Estes resultados demonstram que o albendazole alterou a produ??o de espermatozoides o que interfere da produ??o de ovos de F. hepatica, demonstrando a efic?cia do mesmo neste experimento.
