Resumo: The tick Amblyomma sculptum is known for the fact that its bite causes extremely itchy lesions and for being the transmitting species of the bacterium Rickettsia rickettsii, which causes Brazilian Spotted Fever in humans. Any of its parasitic stages can be found in domestic and wild animals and even in humans, but the capybara is one of its preferred hosts. Due to the similarity between capybaras and guinea pigs, this second species was used to carry out this work, replacing the first one. The general objective of this work was to evaluate the efficacy of fluazuron in the control of the tick A. sculptum in guinea pigs artificially infested with larvae. The work was divided into six stages that occurred chronologically (Study I to VI). In Study I, the influence of three different doses of fluazuron administered by gavage on plasma concentration and efficacy in controlling A. sculptum in guinea pigs was determined. In Study II, the descriptive pharmacokinetics of fluazuron was performed after its administration by gavage to guinea pigs. With the results obtained in the first two studies, in Study III, different bait formulations were developed that underwent evaluations of organoleptic characteristics and resistance to different temperatures and humidity. Fluazuron was incorporated into the chosen formulation, which underwent dose uniformity tests. Subsequently, Study IV was conducted to assess the palatability of the formulation developed for guinea pigs, where baits containing fluazuron (treated group) and placebo group (not treated) were offered. In Study V the baits were administered to guinea pigs to complete the descriptive pharmacokinetics of fluazuron, following the same methodology applied in Study II. In this step, the animals were divided into groups where each received the bait containing fluazuron in a single dose on different pre-infestation experimental days. In Study I, the efficacy results showed that even the lowest dose achieved 100% efficacy against molting engorged larvae. As for plasma bioavailability, there was an increase in the plasma concentration of fluazuron with the increase in dose on day +1. However, from day +4 to the last day of evaluation (+21), mean plasma concentrations did not differ significantly between groups. In Studies II and V, fluazuron plasma concentrations increased rapidly, indicating rapid absorption and slow elimination. The results allowed the development of a bait resistant to different temperatures and humidity, which fluazuron was incorporated and approved in the dose uniformity tests (Study III). In Study IV the placebo and treated baits were palatable to guinea pigs at least 92% of the time. In Study VI, the efficacy obtained was 64.99% for the treated animals 21 days before the infestation, 61.88% for the animals treated 14 days before the infestation and 59.31% for the animals treated 7 days before the infestation. Fluazuron was effective in controlling the A. sculptum tick in guinea pigs.

Qualquer uma de suas fases parasit?rias podem ser encontradas em animais dom?sticos, selvagens e at? mesmo no homem, mas a capivara ? um de seus hospedeiros preferenciais. Devido ? semelhan?a da capivara com porquinhos-da-?ndia, esta segunda esp?cie foi utilizada para a execu??o desse trabalho em substitui??o ? primeira. O objetivo geral desse trabalho foi avaliar a efic?cia do fluazuron no controle do carrapato A. sculptum em porquinhos-da-?ndia infestados artificialmente com larvas do carrapato. Para isso, o trabalho foi dividido em seis etapas que ocorreram de forma cronol?gica (Estudo I ao VI). No Estudo I foi determinada a influ?ncia de tr?s diferentes doses de fluazuron administrado por gavagem na concentra??o plasm?tica e na efic?cia no controle de A. sculptum em porquinhos-da-?ndia. No Estudo II foi realizada a farmacocin?tica descritiva do fluazuron ap?s sua administra??o por gavagem para porquinhos-da-?ndia. Com os resultados obtidos nos dois primeiros estudos, no Estudo III, foram desenvolvidas diferentes formula??es em isca que passaram por avalia??es das caracter?sticas organol?pticas e de resist?ncia ? diferentes temperaturas e umidades. O fluazuron foi incorporado na formula??o escolhida, que passou por testes de uniformidade de dose. Em seguida, foi realizado o Estudo IV para avaliar a palatabilidade da formula??o desenvolvida para porquinhos-da-?ndia, onde foram oferecidas iscas contendo (grupo tratado) e n?o contendo (grupo placebo) fluazuron. No Estudo V as iscas foram administradas para porquinhos-da-?ndia para determinar a farmacocin?tica descritiva do fluazuron, seguindo a mesma metodologia aplicada no Estudo II. Por fim, a ?ltima etapa (Estudo VI), determinou a efic?cia da isca contendo fluazuron administrada para porquinhos-da-?ndia no controle do carrapato A. sculptum. Nesta etapa, os animais foram divididos em grupos onde cada um recebeu a isca contendo fluazuron em dose ?nica em dias experimentais pr?-infesta??o distintos. No Estudo I, os resultados de efic?cia mostraram que mesmo a dose mais baixa alcan?ou 100% de efic?cia contra a muda das larvas ingurgitadas. Quanto a biodisponibilidade plasm?tica, ocorreu um aumento na concentra??o plasm?tica de fluazuron com o aumento da dose no dia +1. No entanto, do dia +4 at? o ?ltimo dia de avalia??o (+21), as concentra??es plasm?ticas m?dias n?o diferiram significativamente entre os grupos. Nos Estudos II e V, as concentra??es plasm?ticas de fluazuron aumentaram rapidamente, indicando absor??o r?pida e elimina??o lenta. Os resultados permitiram o desenvolvimento de uma isca resistente ? diferentes temperaturas e umidade, a qual o fluazuron foi incorporado e aprovado nos testes de uniformidade de dose (Estudo III). No Estudo IV as iscas placebo e tratado foram palat?veis para porquinhos-da-?ndia em, pelo menos, 92% das vezes. A efic?cia obtida das iscas foi 64,99% para os animais tratados 21 dias antes da infesta??o, de 61,88% para os animais tratados 14 dias antes da infesta??o e de 59,31% para os animais tratados 7 dias antes da infesta??o. description The tick Amblyomma sculptum is known for the fact that its bite causes extremely itchy lesions and for being the transmitting species of the bacterium Rickettsia rickettsii, which causes Brazilian Spotted Fever in humans. Any of its parasitic stages can be found in domestic and wild animals and even in humans, but the capybara is one of its preferred hosts. Due to the similarity between capybaras and guinea pigs, this second species was used to carry out this work, replacing the first one. The general objective of this work was to evaluate the efficacy of fluazuron in the control of the tick A. sculptum in guinea pigs artificially infested with larvae. The work was divided into six stages that occurred chronologically (Study I to VI). In Study I, the influence of three different doses of fluazuron administered by gavage on plasma concentration and efficacy in controlling A. sculptum in guinea pigs was determined. In Study II, the descriptive pharmacokinetics of fluazuron was performed after its administration by gavage to guinea pigs. With the results obtained in the first two studies, in Study III, different bait formulations were developed that underwent evaluations of organoleptic characteristics and resistance to different temperatures and humidity. Fluazuron was incorporated into the chosen formulation, which underwent dose uniformity tests. Subsequently, Study IV was conducted to assess the palatability of the formulation developed for guinea pigs, where baits containing fluazuron (treated group) and placebo group (not treated) were offered. In Study V the baits were administered to guinea pigs to complete the descriptive pharmacokinetics of fluazuron, following the same methodology applied in Study II. In this step, the animals were divided into groups where each received the bait containing fluazuron in a single dose on different pre-infestation experimental days. In Study I, the efficacy results showed that even the lowest dose achieved 100% efficacy against molting engorged larvae. As for plasma bioavailability, there was an increase in the plasma concentration of fluazuron with the increase in dose on day +1. However, from day +4 to the last day of evaluation (+21), mean plasma concentrations did not differ significantly between groups. In Studies II and V, fluazuron plasma concentrations increased rapidly, indicating rapid absorption and slow elimination. The results allowed the development of a bait resistant to different temperatures and humidity, which fluazuron was incorporated and approved in the dose uniformity tests (Study III). In Study IV the placebo and treated baits were palatable to guinea pigs at least 92% of the time. In Study VI, the efficacy obtained was 64.99% for the treated animals 21 days before the infestation, 61.88% for the animals treated 14 days before the infestation and 59.31% for the animals treated 7 days before the infestation. Fluazuron was effective in controlling the A. sculptum tick in guinea pigs.
