Resumo: Vitamin A is essential to the human beings, being ingested through animal sources as meat and eggs, or gotten for the conversion of pro vitamin carotenoids found mainly in fruits and vegetables. The vitamin A deficiency is the major nutritional problem that affects underdeveloped populations. As a consequence of this deficiency, xerophthalmia, blindness and premature death can occur, mainly in children. Vitamin A is also important for growth maintenance, for reproductive efficiency, maintenance of the epithelial tissues and keratinization prevention, beyond presenting an important role into the immunological response. Nowadays, the deficient intake of vitamin A is the major nutritional problem found into developing countries populations or poor population of the developed countries. Around of 4.4 million of children, within preschool age, are presenting vision damages due to the deficiency of this vitamin, every year about 250-500 thousand children become blind. Carotenoids are the most important source of retinol for populations in which the consumption of this vitamin is low. For this reason, quantitative and qualitative analysis of carotenoids in foods is very important. In this work, a specific chromatographic methodology was developed to separate the carotenoids and their isomers. Chromatographic separation of the main food carotenoids was optimized and validated. The chromatographic standards were isolated with appropriated amount and purity, and their stability was evaluated to determine the better conditions of storage, as well as shelf life. The isolation of all chromatographic standards was carried out with open column, quantification was done using spectrophotometry and purity evaluation was measured by HPLC. Many matrices have been evaluated as a source of standards, some new sources were found and showed better characteristics, as purity higher than 90%, if compared with those, frequently, used for this purpose. The study of stability showed that mixtures of carotenoids used as external standard for calibration can be storage more than 370 days. Validation study showed that this methodology of analysis has good selectivity, sensitivity and linearity in a concentration range from 0.5 to 8.0 ?g/mL for each carotenoid.

A defici?ncia de vitamina A (retinol) ? o maior problema nutricional em popula??es subdesenvolvidas e suas conseq??ncias s?o a xeroftalmia, cegueira e morte prematura, especialmente em crian?as. A vitamina A ? tamb?m importante na manuten??o do crescimento, na efici?ncia reprodutiva, na manuten??o dos tecidos epiteliais e na preven??o de queratiniza??o, al?m de apresentar uma importante a??o na resposta imunol?gica. A ingest?o deficiente de vitamina A, especialmente em popula??es de pa?ses subdesenvolvidos ou popula??es pobres de pa?ses desenvolvidos, ? o maior problema nutricional da atualidade. Cerca de 4,4 milh?es de crian?as em idade pr?-escolar apresentam danos ? vis?o devido ? defici?ncia desta vitamina e anualmente 250 a 500 mil dessas crian?as acabam cegas. Os caroten?ides s?o a fonte de retinol mais importante em popula??es onde a ingest?o dessa vitamina ? baixa. Desta maneira, a sua correta an?lise, tanto qualitativa quanto quantitativa, em alimentos ? muito importante. Neste trabalho, desenvolveu-se uma metodologia cromatogr?fica espec?fica para a separa??o dos caroten?ides e seus is?meros. Este estudo prop?s a otimiza??o da separa??o cromatogr?fica dos principais caroten?ides encontrados em alimentos e a posterior valida??o do m?todo implantado. Para isto foi necess?rio o isolamento dos padr?es desses caroten?ides em quantidades e purezas suficientes. Uma avalia??o da estabilidade dos padr?es preparados foi realizada para determinar as melhores condi??es de armazenamento, assim como o per?odo m?ximo de armazenamento. O isolamento dos padr?es deu-se por cromatografia aberta com posterior quantifica??o espectrofotom?trica e determina??o da pureza atrav?s de CLAE. Avaliaram-se diversas matrizes naturais como fonte dos caroten?ides de interesse e foram descobertas novas matrizes com caracter?sticas melhores que as usuais para o isolamento desses padr?es, produzindo padr?es com purezas cromatogr?ficas superiores ? exig?ncia m?nima de 90%. O estudo de estabilidade mostrou que ? poss?vel o armazenamento dos padr?es, em misturas multi padr?es prontas para o uso para calibra??o cromatogr?fica externa, por mais de 370 dias.
