This work describes the isolation and structural determination of special metabolites isolated from of the wood of Simira grazielae species (Rubiaceae). This is the first phytochemical study of a specimen of S. grazielae. The material for study was collected in the Reserva Florestal do Vale do Rio Doce, Linhares- ES. The compounds described in this phytochemistry investigation were isolated by solvents partition with n-hexane (C6H14), dichloromethane (CH2Cl2) and ethyl acetate (EtAcO) and classical chromatographyc techniques of the extracts obtained by maceration at room temperature with methanol. Chromatographic fractionation and analysis of the fractions led to the isolation and identification of 18 substances and 1 derivative. Among the special metabolites are isolated: a mixture of hydrocarbons; a mixture of unsaturated hydrocarbons, a mixture of B-stosterol and stigmasterol, 3-O-B-D-glucopyranosyl sitosterol; the phenolic compound 3,4,5-trimetoxifenol and -6'-O-vaniloiltachioside; the aldeide syringaldehyde; the lignan syringaresinol; the coumarins isofraxidine (7-hydroxy-6,8-dimethoxycoumarin), escopoletina (7-hydroxy-6-B-methoxycoumarin), umbelliferone (7-hydroxycoumarin), trimethoxycoumarin and dimethoxycoumarin and the alkaloids B?carboline harman (1-methyl-9H-pyrido[3,4-B]indole, chemotaxonomic character of the genus), ofiorine alkaloids and N-acetyl serotonin . The structural determination was based on the 1H and 13C NMR (1D and 2C) and ESI-HRMS and GC-MS analysis. The MeOH extract and fractions obtained by partition with organic solvents were submitted to evaluation of antioxidant activity with DPPH and potential antinociceptive, revealing expressive activities.
