Filogeografia molecular de poaia (Psychotria ipecacuanha (Brot.) Stokes)

Resumo: Psychotria ipecacuanha is a medicinal species of the Rubiaceae family, popularly known as poaia, which grows in perennial aggregates denominated clumps, in moist and shady areas of the tropical forests of the American continent. The present work aimed at an investigation of the phylogeography of Psychotria ipecacuanha. The sequence of the chloroplast gene trnT-trnL from 40 individuals of 15 natural populations was obtained for this purpose. Nine of these populations were identified in the southeast and six in the central-western area of Brazil. A total of 959 bases were obtained by the sequence data. These aligned sequences showed the existence of 13 polymorphic sites among them, which differentiated eight haplotypes. All individuals sampled in the central-western area had a single haplotype, while the individuals of the southeast area were each allocated in one of the seven remaining haplotypes. The analysis of the data via nested clade analyses made the achievement of a single net possible and it was inferred that the components of this net are grouped at the level of a total cladogram in agreement with the geographical area of origin. The separation among the populations of the southeast and of the center-west follows a pattern that suggests allopatric fragmentation, while certain populations limited to the southeast suffer restriction of gene flow through isolation by distance. In the elaboration of a program of genetic conservation, the populations from the Rio Doce State Park, from Itaperuna, and from Guaraciaba can be indicated as priority because besides presenting variation within the haplotype number of the population, they also have five private haplotypes. Furthermore, these three populations together hold 75% of the variability of the lineages of the trnT-trnL gene.

O presente trabalho visou investigar a filogeografia da poaia. Para isto, seqüenciou-se o gene cloroplastídico trnT-trnL de 40 indivíduos originários de 15 populações naturais. Nove destas populações foram identificadas na região Sudeste e seis na região Centro-Oeste do Brasil. Os dados de seqüenciamento possibilitaram a obtenção de um total de 959 bases. O alinhamento destas seqüências mostrou a existência de 13 sítios polimórficos entre elas, que diferenciaram oito haplótipos. Todos os indivíduos amostrados na região Centro-Oeste possuíam um único haplótipo, enquanto cada um dos indivíduos da região Sudeste foi alocado em um dos sete haplótipos restantes. A análise dos dados via nested clade analyses possibilitou a obtenção de uma rede única e se inferiu que os componentes desta rede estão agrupados em nível de cladograma total, de acordo com a região geográfica de procedência. A separação entre as populações do Sudeste e do Centro-Oeste segue um padrão que sugere fragmentação alopátrica, enquanto certas populações restritas ao Sudeste estão sofrendo restrição ao fluxo gênico por isolamento à distância. Na elaboração de um programa de conservação genética, as populações do Parque Estadual do Rio Doce, Itaperuna e Guaraciaba podem ser indicadas como prioritárias, pois além de apresentarem variação no número de haplótipos dentro da população, possuem também cinco haplótipos privativos.
