Resumo: Plants are a great source of study for obtaining new substances with biological activities. In this context, due harmful adverse effects of current anti-inflammatory drugs, there is a need to identify new alternative substances. In recent studies by our research group, 47 different species of the Fabaceae family were evaluated for their anti-inflammatory potential, among which Acacia polyphylla showed significant results of anti-inflammatory activity in the ex vivo assay and also in the inhibition of edema of the ear in vivo. As a result, this work was intended to carry out a phytochemical study of the species A. polyphylla, contributing to the identification of new compounds responsible for the anti-inflammatory activity. For this, using traditional chromatographic techniques, several fractions of the A. polyphylla were obtained, which were evaluated in an ex vivo anti-inflammatory assay, using human blood. Among the sample tested, it was observed that the BH fraction had the best percentage of PGE2 inhibition (74.8%) when compared to the reference drugs dexamethasone and indomethacin (90.5 and 98.4%, respectively), demonstrating an significant potential for anti-inflammatory activity. The prostaglandin E2 (PGE2) is one of the main mediators of inflammation produced by the COX-1 and COX-2 enzyme pathways, which increases under inflammatory stimuli, which include infectious processes by bacteria, viruses, fungi and parasites, tissue necrosis, foreign bodies, reactions immunology, among others. Furthermore, part of the crude extract of A. polyphylla was analyzed by HPLC, allowing the isolation of two compounds, identified as astragalin and an unpublished compound. Another three compounds were isolated on the bench and confirmed by HPLC. In addition, the anti-inflammatory activity of the isolated compounds was investigated, where astragalin showed a significant PGE2 inhibition activity (48.3%). Through this study, it was possible to contribute to the phytochemical of A. polyphylla, identifying two compounds, including an unpublished compound and astragalin, reported for the first time in this specie, in addition to being able to confirming its anti- inflammatory potential through the ex vivo assay.

Nesse contexto, mediante aos efeitos nocivos que o uso dos anti-inflamatórios trazem, surge a necessidade do estudo de novas substâncias alternativas. Em estudos recentes do nosso grupo de pesquisa, avaliaram-se 47 espécies diferentes da família Fabaceae quanto ao seu potencial anti-inflamatório, dentre essas a Acacia polyphylla apresentou resultados significativos de atividade anti-inflamatória no ensaio ex vivo e também na inibição de edema de orelha in vivo. Portanto, nesse trabalho pretendeu-se realizar o estudo fitoquímico da espécie A. polyphylla, contribuindo para identificação de novos compostos responsáveis pela atividade anti-inflamatória. Para isso, utilizando-se de técnicas cromatográficas tradicionais obtiveram-se diversas frações da A. polyphylla, que foram avaliadas em um ensaio anti-inflamatório ex vivo, utilizando sangue humano. Entre as frações testadas, observou-se que a fração BH obteve o melhor percentual de inibição de PGE2 (74,8%), se comparada aos fármacos de referência dexametasona e indometacina (90,5 e 98,4 %, respectivamente), demonstrando um bom potencial da atividade anti-inflamatória. A prostaglandina E2 (PGE2) é um dos principais mediadores da inflamação produzidos pelas vias das enzimas COX-1 e COX-2, que aumenta sob os estímulos inflamatórios, que incluem os processos infecciosos por bactéria, vírus, fungos e parasitas, necrose tecidual, corpos estranhos, reações imunológicas entre outros. Ademais, parte do extrato bruto da A. polyphylla foi analisado por HPLC, permitindo o isolamento de dois compostos, identificados como astragalina e um composto inédito. Outros três compostos foram isolados em bancada e confirmados por HPLC. Também foi investigada a atividade anti-inflamatória dos compostos isolados, onde a astragalina apresentou uma atividade significativa de inibição de PGE2 (48.3%).
