Resumo: The study is a constant on wound treatments illness that affects a large part of the world population, ranging from burns to more serious diseases such as diabetes. Thus, a better coating is proposed that replaces epithelial functions without contamination by microorganisms. A very studied biopolymer for this purpose is chitosan. It is derived from chitin and found mainly in exoskeletons of insects and crustaceans. Grape is a fruit rich in phenolic compounds that concentrate mainly on skin, seed and stalks. Both chitosan and grape have healing, anti-inflammatory and antibacterial properties. Thus, through this study were developed and characterized chitosan membranes plus grape extract for potential application in the treatment of cutaneous wounds. A first step of this study was the obtaining and characterization of the extract of the cultivar Jacques (Vitis bourquina), considering the whole berry. The extract was obtained by hydroalcoholic maceration of 50% v / v ethanol. From the ready-made extract, the qualitative and semi-quantitative analyzes were followed, besides the anti-radicular activity, FTIR analysis, so that it was possible to characterize the compounds present, and analysis of the antimicrobial activity. The qualitative and semi-quantitative analyzes indicated the presence of phenolic compounds and condensed tannins in the extract. The FTIR spectra showed the existence of hydroxyl groups belonging to phenolic compounds and water. It is also possible to observe the presence of bands related to aromatic molecules derived from pyran referring to condensed tannins. The extract showed low antiradical activity compared to the quercetin and gallic acid standards, possibly due to the small presence of flavonoids. Regarding the antimicrobial analysis, the concentration of the extract used did not present inhibition halo formation for the analyzed bacteria. In the second stage of the research, the membranes were synthesized and characterized by moisture content, water behavior, solubility, water vapor permeability and also analyzed by FTIR, DSC, SEM and analysis of antimicrobial activity. The membranes containing chitosan and grape extract presented uniformity in composition and transparency, an ideal factor for the maintenance of cutaneous wounds. They also showed satisfactory moisture, water absorption capacity and passage of water vapors, maintained integrity in the solubility test, and also showed thermal resistance above body temperature. Through these analyzes and also by the FTIR expectation, it was possible to verify the interaction between the chitosan and the extract. However, antibacterial activity was not observed on any membrane. It is concluded that the extraction method employed was efficient, because in the extract obtained there are presence of healing compounds of interest, as well as the chitosan membranes added with this extract had satisfactory properties for application in the treatment of cutaneous wounds.

Com isso, busca-se um melhor recobrimento que substitua as funções epiteliais sem que haja contaminações por microrganismos. Um biopolímero muito estudado para este fim é a quitosana. Ela é derivada da quitina e encontrada principalmente em exoesqueletos de insetos e crustáceos. A uva é uma fruta rica em compostos fenólicos que se concentram principalmente na casca, semente e engaços. Tanto a quitosana quanto a uva possuem propriedades cicatrizantes, anti-inflamatórias e antibacterianas. Assim, por meio deste estudo foram desenvolvidas e caracterizadas membranas de quitosana acrescidas de extrato de uva para potencial aplicação no tratamento de feridas cutâneas. Uma primeira etapa deste estudo foi a obtenção e caracterização do extrato da cultivar Jacques (Vitis bourquina), considerando a baga inteira. O extrato foi obtido por maceração hidroalcoolica de etanol 50% v/v. A partir do extrato pronto, seguiu-se para as análises qualitativas e semi-quantitativas, além da atividade antirradicalar, análise de FTIR, para que fosse possível caracterizar os compostos ali presentes, e análise da atividade antimicrobiana. As análises qualitativas e semi-quantitativas apontaram para a presença de compostos fenólicos e taninos condensados no extrato. O espectro de FTIR comprovou a existência de grupos hidroxilas pertencentes a compostos fenólicos e água. Também pode-se observar a presença de bandas relacionadas a moléculas aromáticas derivados de pirano referente a taninos condensados. O extrato mostrou baixa atividade antirradicalar comparada com os padrões quercetina e ácido gálico, possivelmente em razão da pequena presença de flavonoides. Com relação à análise antimicrobiana, a concentração do extrato empregada não apresentou formação de halo de inibição para as bactérias analisadas. Na segunda etapa da pesquisa, as membranas foram sintetizadas e caracterizadas quanto aos teores de umidade, comportamento hídrico, solubilidade, permeabilidade ao vapor d’água e também analisadas pelo FTIR, DSC, MEV e análise da atividade antimicrobiana. As membranas contendo quitosana e extrato de uva apresentaram uniformidade em sua composição e transparência, fator ideal para a manutenção das feridas cutâneas. Elas também apresentaram umidade, capacidade de absorção de água e passagem de vapores de água satisfatórios, mantiveram a integridade no teste de solubilidade, e ainda, mostraram resistência térmica acima da temperatura corpórea. Por meio destas análises e também pelo expectro de FTIR foi possível averiguar a interação entre a quitosana e o extrato. Entretanto, a atividade antibacteriana não foi observada em nenhuma membrana.
