Resumo: The genus Garcinia also known as Rheedia, is the biggest of the family Clusiaceae and rich in a considerable variety of compounds such as benzophenones polipreniladas, flavonoids, and xanthones proanthocyanins. The species Garcinia brasiliensis is a plant native to the Amazon region and its fruits are popularly known as "bacupari". In this work, from the ethyl acetate extract of the seeds of Garcinia brasiliensis were isolated the natural poliprenilada benzophenone guttiferone-A. Molecular changes in the phenolic hydroxyl of this compound led to the formation of seven novel semisynthetic derivatives, which were evaluated for anti-proteolytic activity, through experiments on the enzymes trypsin and papain. The interest in the search of protease inhibitors is justified by the proven link between this class of enzymes to a variety of diseases, including among others, cancer and AIDS. The guttiferone-A, showed significant results for the two enzymes tested, demonstrating the importance of the prenyl and hydroxyl groups of the molecule. Furthermore, the acylated derivatives also showed a significant IC50 compared to original product, especially two of them, is still possible to relate the inhibitory activity of these molecules in the presence of carbonyl group.

A espécie Garcinia brasiliensis é uma planta nativa da região amazônica e seus frutos são popularmente conhecidos como “bacupari”. Neste trabalho, a partir do extrato acetato de etila das sementes de Garcinia brasiliensis, foi possível isolar a benzofenona natural poliprenilada guttiferona-A. Modificações moleculares nas hidroxilas fenólicas deste composto levaram a obtenção de sete derivados semissintéticos inéditos, os quais foram avaliados quanto à atividade antiproteolítica, através de ensaios sobre as enzimas tripsina e papaína. O interesse pela busca de inibidores de proteases justifica-se pela comprovada relação desta classe de enzimas a uma série de patologias, incluindo entre outras, o câncer e a AIDS. A guttiferona-A, apresentou resultados relevantes para as duas enzimas testadas, demonstrando a importância dos grupos prenila e hidroxila da molécula.
