Resumo: | There is a constant increase in the search for quality of life, which has led the population to develop healthier habits. Oils and fats, very present in the routine and basic part of a diet, have been paying attention, and when consumed in excess they can trigger several diseases. Grape seed oil is a raw material in several branches in the industry, rich in vitamin E, antioxidants and essential oils, corroborating the importance of developing technologies that disseminate its use to take advantage of its benefits. In view of this, dietary triglycerides have stood out due to their nutraceutical properties, lower caloric value, greater solubility and easier metabolism in the body, compared to conventional ones. Despite its many benefits, the use of enzymes may not be advantageous, as it increases costs in the process. A very promising alternative is the immobilization of the enzyme, since this method allows the reuse of the biocatalyst. For these processes, fixed bed reactors (PBR) are commonly used, due to their efficiency and high production. In this work, the objective was to synthesize dietary triglycerides of the MLM type in acidolysis reaction, using grape seed oil and capric acid (C10), catalyzed by Rhizopus oryzae lipase immobilized by physical adsorption in corn cob powder. The immobilized biocatalyst was characterized in terms of optimum temperature and pH using an experimental central rotational composite design (DCCR). It was also evaluated the thermal stability, as well as the kinetic and thermodynamic parameters of the free and immobilized enzyme, at different temperatures. Through experimental design, central rotational composite design (DCCR), the molar ratio, temperature and amount of biocatalyst suitable for a higher degree of incorporation (GI) were determined. Finally, the synthesis was performed in a fixed bed reactor and the product was analyzed for GI and triglyceride percentages. The results obtained allowed the selection of the most efficient reaction conditions for obtaining dietary triglycerides of the MLM type, with promising results in biotechnology. |

Óleos e gorduras, muito presentes na rotina e parte base de uma alimentação têm tomado atenção, e quando consumidos em excesso podem desencadear diversas doenças. O óleo de semente de uva é matéria prima em vários ramos na indústria, rico em vitamina E, antioxidantes e óleos essenciais, corroborando à importância de se desenvolver tecnologias que disseminem seu uso para aproveito de seus benefícios. Visto isso, os triglicerídeos dietéticos vêm se destacando devido suas propriedades nutracêuticas, menor valor calórico, maior solubilidade e metabolização facilitada no organismo, comparado aos convencionais. Apesar de seus diversos benefícios, o uso de enzimas pode não ser vantajoso, por elevar custos no processo. Uma alternativa bastante promissora é a imobilização da enzima, já que esse método permite a reutilização do biocatalisador. Para esses processos, os reatores em leito fixo (PBR) são comumente utilizados, devido a sua eficiência e alta produção. Neste trabalho, o objetivo foi sintetizar triglicerídeos dietéticos do tipo MLM em reação de acidólise, utilizando óleo de semente de uva e ácido cáprico (C10), catalisada por lipase de Rhizopus oryzae imobilizada por adsorção física em pó de sabugo de milho. O biocatalisador imobilizado foi caracterizado quanto à temperatura e pH ótimos empregando-se um planejamento experimental de delineamento composto central rotacional (DCCR). Também se avaliou a estabilidade térmica, bem como os parâmetros cinéticos e termodinâmicos da enzima livre e imobilizada, em diferentes temperaturas. Através de planejamento experimental, delineamento composto central rotacional (DCCR), foram determinados a razão molar, temperatura e quantidade de biocatalisador adequados para maior grau de incorporação (GI). Por fim, a síntese foi realizada em reator de leito fixo e o produto analisado quanto o GI e porcentagens de triglicerídeos. Os resultados obtidos permitiram a seleção das condições reacionais mais eficientes para obtenção dos triglicerídeos dietéticos do tipo MLM, com resultados promissores na área da biotecnologia.application/pdfporUniversidade Federal de AlfenasPrograma de Pós-Graduação em Engenharia QuímicaUNIFAL-MGBrasilInstituto de Ciência e Tecnologiainfo:eu-repo/semantics/openAccesshttp://creativecommons.org/licenses/by-nc-nd/4.0/Triglicerídeos.Lipase.Enzimas imobilizadas.Reatores biológicos.ENGENHARIASSíntese de triglicerídeos dietéticos catalisada por lipase de Rhizopus oryzae imobilizada em pó de sabugo de milho por adsorção físicaSynthesis of dietary triglycerides catalyzed by Rhizopus oryzae lipase immobilized on corn cob powder by physical adsorptioninfo:eu-repo/semantics/masterThesisinfo:eu-repo/semantics/publishedVersion-42974172594986389316006004518971056484826825reponame:Repositório Institucional da Universidade Federal de Alfenas - RiUnifalinstname:Universidade Federal de Alfenas (UNIFAL)instacron:UNIFALPacheco, Bruna Jeanne SoaresCC-LICENSElicense_urllicense_urltext/plain;
