Resumo: Adrenaline is released into the blood by the sympathetic nervous system and the suprarenal medulla. It is of great importance for organisms acting from the prolongation of the effect of local anaesthetics to their action on the heart and can restore heart rate in the event of cardiac arrest. In this way, it presents itself as an important topic of study. On the other hand, paperbased biosensors, because they are a relatively simple technique, inexpensive and easily accessible, also have great potential as a tool for analytical chemistry. These utilize an enzyme that can catalyze an oxidation or reduction of the analyte and generate a signal proportional to the concentration thereof. As a biological element of the proposed biosensor, the enzyme Polyphenoloxidase was used. Therefore, in this work, a potentiometric biosensor for adrenaline determinations was constructed. The reference electrode was printed on parchment paper (Filipinho®) using an inkjet printer (Hp 2050®) with the original ink cartridges. The printing ink was modified with Potassium Chloride and Silver Nitrate. The indicator electrode was constructed from the printing of the conductive part, i.e., ink with KCl. The working electrode was obtained from the immobilization of the enzyme in paste form containing graphite and commercial jelly (Dr. Oetker®). The impression of both electrodes originated, therefore, the proposed biosensor. The enzyme was extracted from the yam (Colocasia sp.) With phosphate buffer (pH = 7.0). The best composition was 1000 μL of Enzymatic Crude Extract (EBE) with 500 μL of jelly and 0.330 g of graphite powder. The portable equipment, for potentiometric measurements, consists of a computer interface (Arduino Duo®) that has analogue inputs with 12-bit resolution. To adjust the input impedance, a circuit (Op-Amp CI OP07, Analog Devices®) of the buffer type was constructed. Communication with the user was done through a program developed in visual language Basic®; the interface was connected to the computer through the USB port. Employing Adrenaline solutions in phosphate buffer (pH = 7.0). The Detection Limit (LD = 2.15 x 10-4 mol L -1) was calculated and the linear response range was 5x 10-2 to 5x10-4 mol L -1. The accuracy was checked against a titration method, and the results were concordant at a 95% level (paired t-test). The system was applied for the determination of adrenaline in injectable drugs, and the results were satisfactory the precision expressed in coefficient of variation was always less than 22%.

The system was applied for the determination of adrenaline in injectable drugs, and the results were satisfactory the precision expressed in coefficient of variation was always less than 22%.A adrenalina é liberada no sangue pelo sistema nervoso simpático e pela medula supra renal, tem grande importância para organismos agindo desde o prolongamento do efeito dos anestésicos locais até sua atuação no coração, podendo restaurar os batimentos cardíacos no caso de uma parada cardíaca. Dessa maneira, ela se apresenta como importante tema de estudo. Por outro lado, os biossensores impressos em papel, por se tratarem de uma técnica relativamente simples, de baixo custo e de fácil acesso, também apresentam grande potencial como ferramenta de química analítica. Esses utilizam uma enzima que pode catalisar uma oxidação ou redução do analito e gerar um sinal proporcional à concentração do mesmo. Como elemento biológico, do biossensor proposto, foi utilizado a enzima Polifenoloxidase. Portanto, neste trabalho foi construído um biossensor potenciométrico para determinações de adrenalina. O eletrodo de referência foi impresso em papel vegetal (Filipinho®) utilizando impressora jato de tinta (Hp 2050®) com os cartuchos de tintas originais. A tinta para impressão foi modificada com Cloreto de Potássio e Nitrato de Prata. O eletrodo indicador foi construído a partir da impressão da parte condutora, isto é, tinta com KCl. O eletrodo de trabalho se deu a partir da imobilização da enzima em forma de pasta contendo grafite e gelatina comercial (Dr. Oetker®). A impressão de ambos os eletrodos originou, portanto, o biossensor proposto. A enzima foi extraída do inhame (Colocasia sp.) com tampão fosfato (pH =7,0). A melhor composição foi de 1000 μL de Extrato Bruto Enzimático (EBE) com 500 μL de gelatina e 0,330 g de pó de grafite. O equipamento portátil, para as medidas potenciométricas, consiste de uma interface computacional (Arduino Duo®) que possui entradas analógicas com resolução de 12 bits. Para ajuste de impedância de entrada construiu-se um circuito (Op-Amp CI OP07, Analog Devices®) do tipo buffer. A comunicação com o usuário foi feita através de um programa desenvolvido em linguagem visual Basic®, a interface foi conectada ao computador através da porta USB. Empregando soluções de Adrenalina em tampão fosfato (pH = 7,0). Foi calculado o Limite de Detecção (LD= 2,15 x 10-4mol L-1) e a faixa de resposta linear foi de 5x 10-2 a 5x10-4 mol L-1. A exatidão foi checada comparando com método titulométrico e houve concordância entre os resultados a um nível de 95 % (teste-t pareado). O sistema foi aplicado para a determinação de adrenalina em medicamentos injetáveis e os resultados foram satisfatório a precisão expressa em coeficiente de variação foi sempre inferior a 22%.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPESapplication/pdfporUniversidade Federal de AlfenasPrograma de Pós-Graduação em QuímicaUNIFAL-MGBrasilInstituto de Químicainfo:eu-repo/semantics/openAccesshttp://creativecommons.org/licenses/by-nc-nd/4.0/AdrenalinaBiossensoresPotenciometriaQUIMICA ANALITICA::INSTRUMENTACAO ANALITICAConstrução de um biossensor potenciométrico impresso para determinação de adrenalinainfo:eu-repo/semantics/masterThesisinfo:eu-repo/semantics/publishedVersion132825307882678230660060060011432645100632034522075167498588264571reponame:Biblioteca Digital de Teses e Dissertações da UNIFALinstname:Universidade Federal de Alfenas (UNIFAL)instacron:UNIFALOliveira, Andressa Cristina Vasques DeLICENSElicense.txtlicense.txttext/plain;
