Resumo: Some transition metals are of great importance in the biological environment, since they interact directly with the biomolecules essential for the maintenance of life. One characteristic of the metals that make them so important as functional and structural components of living things is their propensity to lose electrons easily by forming ions with positive charges, which tend to be soluble in biological fluids. Many studies have considered the possible interactions of metal ions with biomolecules present in the living organism. The present work had as objective the preparation of new compounds of palladium (II), containing iminic ligands, with the intention of obtaining novel compounds that present pharmacological properties. The iminic ligand (Schiff's Base) was prepared by the reaction between the trans-cinnamaldehyde aldehyde and the amine p-aminoacetophenone (Hfafe) L1. The imine ligand obtained was complexed to palladium(II), leading to substitutions, with the aim of producing new coordination compounds of the formula [PdX2(Hfafe)2], where X = Cl (C1), N3 (C2), SCN (C3), Br (C4) and I (C5). The compounds were characterized by melting point or decomposition, solubility, molar conductivity, elemental analysis (CHN), IR spectroscopic techniques, 1H and 13C NMR, thermogravimetric TG-DTA studies, X-ray diffraction for monocrystal of C2 and calculations of DFT of L1. The compounds were used in biological tests for MCF-7 and MDA-MB-435 breast cancer cells, Leishmania ssp, in vitroexperiments of promastigotes and amastigotes L. (L.) brazilienses, antifungal in yeasts of the genus Candida spp.: C. albicans, C. tropicalis, C. krusei, C. glabrata, C. parapsilosis. The free ligand was shown to have a significant effect against biological tests comparable to those of the positive controls. However, the synthesized complexes showed better potential pharmacological agents than their precursor ligand. Among the synthesized complexes, the compounds [Pd(N3)2(Hfafe)2] deserve to be highlighted, which presented, qualitatively, activity against mammary cells, Leishmania and fungi tested.

Uma característica dos metais que os tornam tão importantes como componentes (funcionais e estruturais) dos seres vivos é sua propensão em perder elétrons facilmente formando íons com cargas positivas, que tendem a ser solúveis em fluídos biológicos. Muitos estudos tomaram como base as possíveis interações dos íons metálicos com as biomoléculas presentes no organismo vivo. O presente trabalho teve como objetivo a preparação de novos compostos de paládio(II), contendo ligantes imínicos, com o intuito de obter compostos inéditos que apresentem propriedades farmacológicas. O ligante imínico (Base de Schiff) foi preparado através da reação entre o aldeído trans-cinamaldeído e a amina p-aminoacetofenona (Hfafe) L1. O ligante imínico obtido foi complexado ao paládio(II), conduzindo substituições, com o intuito de se produzir novos compostos de coordenação de fórmula geral [PdX2(Hfafe)2], sendo X = Cl (C1), N3 (C2), SCN (C3), Br (C4) e I (C5). Os compostos foram caracterizados através das técnicas de ponto de fusão ou decomposição, solubilidade, condutividade molar, análise elementar (CHN), das técnicas espectroscópicas de infravermelho (IV), ressonância magnética nuclear (RMN) de 1H e 13C, estudos termogravimétricos TG-DTA, difração de raios-X para o monocristal de C2 e cálculos de DFT de L1. Os compostos foram empregados em testes biológicos, para as células MCF-7 e MDA-MB-435 do câncer de mama, Leishmania ssp, em experimentos in vitro de células promastigotas e amastigotas L. (L.) brazilienses, antifúngico em leveduras do gênero Candida spp.: Candida albicans, Candida tropicalis, Candida krusei, C. glabrata, C. parapsilosis. Verificou-se que o ligante livre apresentou efeito significativo frente os testes biológicos comparáveis aos dos controles positivos. No entanto, os complexos sintetizados se mostraram melhores potenciais agentes farmacológicos do que seu ligante precursor. Entre os complexos sintetizados merece destaque o compostos [Pd(N3)2(Hfafe)2], que apresentou, qualitativamente, atividade frente as células mamárias, a Leishmania e aos fungos testados.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPESapplication/pdfporUniversidade Federal de AlfenasPrograma de Pós-Graduação em QuímicaUNIFAL-MGBrasilInstituto de Químicainfo:eu-repo/semantics/openAccesshttp://creativecommons.org/licenses/by-nc-nd/4.0/Síntese orgânicaPaládio – uso terapêuticoAnálise espectralQuímica bioinorgânicaQUIMICA::QUIMICA INORGANICASíntese, caracterização e avaliação da atividade biológica de complexos de paládio(II) contendo ligante imínicoinfo:eu-repo/semantics/doctoralThesisinfo:eu-repo/semantics/publishedVersion132825307882678230660060060031391270652389310962075167498588264571reponame:Repositório Institucional da Universidade Federal de Alfenas - RiUnifalinstname:Universidade Federal de Alfenas (UNIFAL)instacron:UNIFALGois, Elba Pereira DeLICENSElicense.txtlicense.txttext/plain;
