Resumo: The presence of legumes can influence the diversity and size of the native rhizobial community of the soil. In this context, the objective of this study was to evaluate the effect of the legume and its distance in relation to the rhizobia diversity, in pasture soil of Brachiaria decumbens Stapf. Under silvopastoral system. The soil was collected in an Itambé-PE experiment, with 3 treatments, consisting of 1 ha each, being: Gliricidia + braquiária, Sabiá + braquiária and, in monoculture, with 3 replicates. In the intercropped plots, sampling was done by dividing the same in transects in the direction of the longest length, along which points were scored to obtain the composite sample of soil between the double row of the legume (0 m), and from that plot Point, every four meters (4m, 8m), the latter coinciding in the center of the brachiaria. Already in the parcels with brachiaria in monoculture, the collection was carried out in three random points. The number of soil rhizobia was estimated using the most probable number method in plants, with three replications, in the greenhouse and using the cowpea (Vigna unguiculata (L.) Walp) as bait plant. Germinated seeds were inoculated with 1.0 ml of soil samples after serial dilution (10 -1 to 10 -8) and collected at 35 days. Twenty nodules per treatment were used for isolation and phenotypic characterization. Grouping with groups of 100% similarity was carried out. Random representatives were selected to evaluate the symbiotic capacity together with strains recommended for cowpea, with nitrogen and absolute control. The dry mass of dry nodules (MSN), number of nodules (NN), and relative efficiency (ER) were analyzed. For the sequencing, the amplified DNA of the isolates that showed relative efficiency, similar to or greater than the reference control, was used. Rhizobian populations were found for cowpea in all combinations of legumes and distances, as well as in single brachiaria pasture. The morphological diversity of diazotrophic bacterial communities was high. Significant differences were found for number of nodules, root dry mass, shoot dry mass and relative efficiency. 51% of the isolates showed similar or higher relative efficiency than the positive controls. Most of the isolates presented genetic similarity ranging from 77% to 99% with Bradyrhizobium strains. Therefore, it is concluded that the presence of the tree legumes Gliricídia and Sabiá under silvipastoral system in pastures of B. decumbens Stapf. Do not influence the density and diversity of noduliferous bacteria in cowpea. And that native populations of noduliferous bacteria in cowpea are efficient at fixing nitrogen and can be recommended for tests of agronomic efficiency.

Nesse contexto, o objetivo deste estudo foi avaliar o efeito da leguminosa e sua distância em relação a diversidade rizobiana, em solo de pastagem de Brachiaria decumbens Stapf. sob sistema silvipastoril. O solo foi coletado em experimento instalado em Itambé-PE, com 3 tratamentos, constituídos por parcelas de 1 ha cada, sendo eles: Gliricidia + braquiária, Sabiá + braquiária e, braquiária em monocultivo, com 3 repetições. Nas parcelas com consórcio a amostragem foi feita dividindo-se a mesma em transectos no sentido do maior comprimento, ao longo do qual foram marcados pontos para obtenção da amostra composta de solo entre a fila dupla da leguminosa (0 m), e a partir desse ponto, a cada quatro metros (4m, 8m), coincidindo este último no centro da faixa de braquiária. Já nas parcelas com braquiária em monocultivo, a coleta foi realizada em três pontos aleatórios. A estimativa do número de rizóbios do solo, foi realizada pelo método de número mais provável em plantas, com três repetições, em casa-de-vegetação e utilizando o feijão-caupi (Vigna unguiculata (L.) Walp) como planta isca. Sementes germinadas foram inoculadas com 1,0 ml das amostras de solo, após diluição serial (de 10 -1 a 10 -8) e coletadas aos 35 dias. Foram utilizados 20 nódulos por tratamento para isolamento e caracterização fenotípica. Foi realizado agrupamento, com formação de grupos com 100% de similaridade, dos quais representantes aleatórios foram selecionados para avaliação da capacidade simbiótica conjuntamente com estirpes recomendadas para feijão-caupi, com um controle nitrogenado e outro absoluto. Foram analisadas as variáveis: massa seca da parte aérea (MSPA), massa seca de raiz (MSR), massa seca de nódulos secos (MSN), número de nódulo (NN), e eficiência relativa (ER). Para o sequenciamento foi utilizado o DNA amplificado dos isolados que apresentaram eficiência relativa, semelhante ou maior que as do controle referência. Foram encontradas populações rizobianas para caupi em todas as combinações de leguminosas e distâncias, bem como na pastagem de braquiária solteira. A diversidade morfológica das comunidades bacterianas diazotróficas mostrou-se alta. Foram encontradas diferenças significativas para número de nódulos, massa seca de raiz, massa seca de parte aérea e eficiência relativa. 51% dos isolados apresentaram eficiência relativa semelhantes, ou superiores aos controles positivos. A maioria dos isolados apresentou similaridade genética alcançando de 77% a 99% com linhagens de Bradyrhizobium. Portanto, conclui-se que a presença das leguminosas arbóreas Gliricídia e Sabiá sob sistema silvipastoril em pastagens de B. decumbens Stapf. não influenciam a densidade e diversidade de bactérias nodulíferas em feijão-caupi.
