Effects of environmentally relevant concentrations of the antidepressant sertraline on the performance and behavior of larval amphibians

Resumo: Environmental contamination by pharmaceuticals is receiving an important attention by the scientific community in the last several years. Inadequate disposal, insufficient basic sanitation and technical limitations on sewage treatment results in the detection of several compounds in surface waters around the planet. Antidepressants, particularly Selective Serotonin Recapture Inhibitors, are among them. I conducted two experiments manipulating sertraline, a Selective Serotonin Recapture Inhibitor that is the second most consumed antidepressant in Brazil. The first experiment aimed at evaluating whether environmentally relevant concentrations of sertraline influence the performance of tadpoles of Rhinella icterica as measured by survival, mass gain, development, and time to metamorphosis. No effects were observed on measured response variables. The second experiment aimed at evaluating whether environmental relevant concentrations of sertraline influence the interaction between predators and their prey. My model system comprised tadpoles of the treefrog Scinax fuscovarius as consumers, and larvae of the dragonfly Erythrodiplax sp as predators. As response variables we quantified, in the prey, parameters known to regulate predation risk such as activity level, food consumption, spatial avoidance, refuge seeking, and, swimming kinematics. No main effects of sertraline on behavioral response variables were detected; a significant sertraline-by-predator presence interaction term on activity levels was observed. By contrast, sertraline affected several swimming kinematic variables including time swimming, total displacement and number of stimuli required to induce swimming. Together, these results suggest that environmentally relevant concentrations of sertraline may provoke a reduction in the fitness of amphibian larvae.

Descarte inadequado, saneamento básico insuficiente e limitações técnicas no tratamento de esgoto resultam na detecção de uma variedade de compostos em águas superficiais ao redor do planeta. Entre estes compostos estão os antidepressivos, particularmente aqueles pertencentes à classe dos Inibidores Seletivos da Recaptação de Serotonina. Eu conduzi dois experimentos manipulando sertralina, um inibidor seletivo de recaptação de serotonina que é o segundo antidepressivo mais vendido no Brasil. O objetivo do primeiro experimento foi avaliar os efeitos de concentrações ambientalmente relevantes de sertralina no desempenho de girinos de Rhinella icterica medido através da sobrevivência, ganho de massa, desenvolvimento e tempo para completar a metamorfose. Os resultados indicaram que, dentro das concentrações estudadas, sertralina não provocou nenhum impacto nos parâmetros avaliados. No segundo experimento procurei avaliar se concentrações ambientalmente relevantes de sertralina influenciam a interação entre predadores e presas. O sistema modelo compreendeu girinos da perereca Scinax fuscovarius como consumidores, e larvas da libélula Erythrodiplax como predadores. Como variáveis de resposta quantificamos, na presa, variáveis conhecidas por regular o risco de predação como o nível de atividade, o consumo alimentar, a evitação espacial, a busca por refúgio, e a cinemática de natação. Nossos resultados mostraram que a sertralina não apresentou efeito principal significativo nas variáveis comportamentais; o único efeito comportamental observado foi uma interação significativa entre sertralina e presença de predador para taxa de atividade. Por outro lado, a sertralina afetou diversas variáveis cinemáticas como o tempo de natação, o deslocamento total e o número de estímulos necessários para induzir a natação.
