Resumo: With the onset of pregnancy, profound anatomical and physiological changes affect virtually all organs in the mother’s body. These changes are mainly induced to meet the complex demands of fetal development and protection and to prepare the maternal organism for parturition. MicroRNAs are small non-coding endogenous molecules capable of regulating gene expression in the post-transcriptional period and have been recently studied in order to better comprehend the physiological and pathological conditions of pregnancy. In this sense, the identification of the rs6505162 single nucleotide polymorphism (SNP) is also emphasized. The rs6505162 SNP is located in pre-microRNA-423 and maps to 17q11.2, with a nucleotide change from C to A. This polymorphism has been considered functional, regulating levels microRNA-423 expression. Thus, this study aimed to evaluate the association between rs6505162 SNP and microRNAs (microRNA-423-5p and microRNA-221-3p) expression in pregnancy. Our sample included 173 pregnant. Questionnaires with socio demographic questions were applied to characterize the sample. And rs6505162 SNP genotyping were evaluated by real time PCR. MicroRNA-423-5p, microRNA-221-3p expression were evaluated by quantitative PCR. Bioinformatics analysis was performing using the DIANA Tools software, mirPath v.3. The results shows an association between the CC genotype and microRNA-221-3p (p = 0.015), as well as with allele C and microRNA-423-5p (p = 0.037) and microRNA-221-3p (p = 0.025). The association was maintained after the adjusted linear regression analysis. The C allele carriers have higher levels of microRNA 423-5p and microRNA-221-3p expression in the blood serum. The correlation analysis showed a positive correlation between the microRNAs (p = 0.001). The bioinformatics analysis revealed the estrogen signaling pathway (p = 0.002), and the Raf-1 proto- 7 oncogene, serine/threonine kinase (RAF1) gene. In conclusion, our results show that the rs6505162 SNP influences the peripheral blood expression of microRNA-423-5p and microRNA-221-3p, and that both microRNAs expression are positively correlated in pregnancy. Thus, this SNP and microRNAs interaction could represent an important study target to seek biomarker for physiological and pathological conditions.

Thus, this SNP and microRNAs interaction could represent an important study target to seek biomarker for physiological and pathological conditions. Submitted by Cristiane Chim (cristiane.chim@ucpel.edu.br) on 2023-10-09T19:45:18Z No. of bitstreams: 1 Dissertação - Cainá Corrêa do Amaral.pdf: 1600779 bytes, checksum: b51d0d9c31aec89413e6ac4936289645 (MD5)Made available in DSpace on 2023-10-09T19:45:19Z (GMT).
