São Bernardo do Campo, 2022O córtex insular anterior (AIC) consiste de uma região de integração sensorial. Ele parece detectar eventos salientes para guiar comportamento direcionado a objetivo, codificar erro e estimar a passagem do tempo. O processamento temporal nessa região pode ocorrer pela integração de representações interoceptivas. As projeções entre AIC e córtex pré-frontal medial (mPFC) - observadas tanto em ratos quanto em humanos - também sugerem um possível papel dessas estruturas na integra- ção de respostas autonômicas com comportamento em curso. Poucos estudos, no entanto, investigaram o papel do AIC e do mPFC em tarefas de tomada de de- cisão e estimação temporal. Além disso, os achados não são uniformes, de modo que a relação entre a tomada de decisão temporal e essas áreas permanece incerta. O presente estudo empregou inativações bilaterais para explorar o papel do AIC e pré-límbico (PL) em ratos durante uma tarefa de tomada de decisão temporal. Nesta tarefa, duas alavancas estão disponíveis simultaneamente (mas apenas uma está ativa), uma disponibilizando reforço após um curto intervalo e a outra após um longo intervalo fixo. O desempenho ideal requer uma mudança da alavanca curta para a longa após o intervalo fixo curto ter decorrido e nenhum reforço ter sido liberado. Os achados do Experimento I mostraram que o comportamento do switch da alavanca curta para a longa depende do AIC e PL. Durante a inativação do AIC, as latências de switch tornaram-se mais variáveis, enquanto durante a inativação do PL as latências de switch tornaram-se mais variáveis e menos precisas. Por sua vez, os resultados do Experimento II mostraram que os animais são sensíveis à mudança na probabilidade de apresentação das tentativas longas, ajustando as latências de switch para maximizar o ganho de reforço. A inativação do AIC aumentou as latên- cias de switch na baixa probabilidade e aumentou a variabilidade das latências na baixa e na alta probabilidade. Esses achados apontam para uma dissociação entre AIC e PL na tomada de decisão temporal, sugerindo que o AIC é importante para a precisão temporal, e o PL é importante tanto para acurácia quanto para a preci- são temporal. Este projeto tem relevância no entendimento dos mecanismos neurais envolvidos no processamento de incerteza de recebimento de reforço em função do tempo.The anterior insular cortex (AIC) comprises a region of sensory integration. It appears to detect salient events to guide goal-directed behavior, code tracking errors, and estimate the passage of time. Temporal processing in this region could occur by in- tegrating representations of interoception. Projections between the AIC and medial prefrontal cortex (mPFC) - found both in rats and humans - also suggest a possible role for these structures in the integration of autonomic responses during ongoing behavior. Few studies, however, have investigated the role of AIC and mPFC in decision-making and time estimation tasks. Moreover, their findings are not consis- tent, so the relationship between temporal decision-making and those areas remains unclear. The present study employed bilateral inactivations to explore the role of AIC and prelimbic (PL) in rats during a temporal decision-making task. In this task, two levers are available simultaneously (but only one is active), one predic- ting reinforcement after a short, and the other after a long fixed interval. Optimal performance requires a switch from the short to the long lever after the short fixed interval elapsed and no reinforcement was delivered. The results of the Experiment I showed that switch behavior from the short to the long lever was dependent on AIC and PL. During AIC inactivation, switch latencies became more variable, while du- ring PL inactivation switch latencies became both more variable and less accurate. The results of Experiment II showed that the animals are sensitive to the change in the probability of long trials, adjusting switch latencies in order to maximize the reinforcement. Inactivating the AIC increased switch latencies at low probability and increased the variability of latencies at low and high probability. These findings point to a dissociation between AIC and PL in temporal decision-making, sugges- ting that the AIC is important for temporal precision, and PL is important for both temporal accuracy and precision.
