Resumo: Our objective was to evaluate the cases of PS spiders in the green seaweed C. mexicana (PS-Cm) in models of gastric damage induced by ethanol in mice and its toxic effects. By studying the gastroprotective action, groups of male Swiss mice were treated with PS-Cm (2, 20 or 200 mg / kg, v.o.) and subjected to gastric injury induced by absolute ethanol. Also, the case, also, the involvement of the final pathways of PG and NO. PS-Cm (2, 20 or 200 mg / kg), when administered orally, significantly reduced gas and gastric, and are inhibited by pre-treatment with indomethacin. PS-Cm (200 mg / kg) prevented the reduction of GSH and normalized the high levels of MDA, induced by ethanol. However, PS-Cm (200 mg / kg) are not important for NO2 / NO3 compared to the ethanol group. Pretreatment with L-NAME induced gastric mucosal damage and did not inhibit the gastroprotective effect of PS-Cm (200 mg / kg). To evaluate the possible toxic effects, we used the acute and repeated dose (28 days) toxicity assays with Swiss mice using OECD guidelines 420 and 407. The results obtained are not acute toxicity tests, they showed that PS-Cm (400, 600, 800, 1000 mg / kg) did not cause obvious toxicity. But at a dose of 2000 mg / kg for evidence of toxicity (piloerection, apathy and weight loss) and death (n = 3). There is no repeated dose toxicity test, animals treated with PS-Cm (600 mg / kg) showed no signs of toxicity. However, animals treated with PS-Cm (800 or 1000 mg / kg) showed signs of toxicity and the group (1000 mg / kg) died during the test. No significant difference was observed in relation to the control group for biochemical analyzes. In the histopathological evaluation, lesions were observed without border and without animals of the groups (800 mg / kg). Regeneration signals were observed in the post-test treated group. Thus we can conclude that the gastroprotective effect of PS-Cm observed to develop the involvement of PG and a reduction of oxidative stress, with actions independent of NO. And, which is a single oral administration of PS-Cm showed a median lethal dose level (LD50) of 1000 mg / kg body weight, being classified in category 3 GHS and that NOAEL was determined to be a dose lower than 600 mg / kg / day when administered orally.

Para se estudar a ação gastroprotetora, grupos de camundongos Swiss machos foram tratados com PS-Cm (2, 20 ou 200 mg/kg, v.o.) e submetidos a lesão gastrica induzidos por etanol absoluto. Avaliou-se, também, o envolvimento das vias endógenas de PG e NO. Os PS-Cm (2, 20 ou 200 mg/kg), quando administrados por via oral, reduziram significativamente o dano gástrico e este efeito foi inibido através do pré-tratamento com indometacina. PS-Cm (200 mg/kg) impediu a redução da GSH e normalizou os níveis elevados de MDA, induzidos pelo etanol. No entanto, os PS-Cm (200 mg/kg) não mostraram efeitos significativos sobre o NO2/NO3 em comparação com o grupo etanol. O pré-tratamento com L-NAME induziu danos da mucosa gástrica e não inibiu o efeito gastroprotetor de PS-Cm (200 mg/kg). Para avaliar os possiveis efeitos tóxicos, utilizamos os ensaios de toxicidade aguda e de dose repetida (28 dias) com camundongos Swiss, utilizando as diretrizes 420 e 407 da OECD. Os resultados obtidos no ensaio de toxicidade aguda, demosntraram que os PS-Cm (400, 600, 800, 1000 mg/kg) não ocasionaram toxicidade evidente. Mas, para a dose de 2000 mg/kg foi evidenciado sinais de toxicidade (piloereção, apatia e perda de peso) e morte (n=3). No ensaio de toxicidade de dose repetida, os animais tratados com PS-Cm (600 mg/kg) não demonstraram sinais de toxicidade. No entanto, os animais tratados com PS-Cm (800 ou 1000 mg/kg) apresentaram sinais de toxicidade e os do grupo (1000 mg/kg) morreram durante o ensaio. Não foi observada diferença significativa em relação ao grupo controle para análises bioquímicas. Na avaliação histopatólogica, lesões foram observadas no rim e no fígado de animais do grupo (800 mg/kg). Os sinais de regeneração foram observados no grupo tratado pós-ensaio. Assim podemos concluir que o efeito gastroprotector de PS-Cm observado se deve ao envolvimento de PG e a redução do estresse oxidativo, com ações independentes do NO.
