Resumo: | This work is divided into two chapters and aims to: 1) Identify the species of ticks in the freeliving phase, which occur in the Conservation Units (UC's) in the states of Rio de Janeiro, Esp?rito Santo (ES) and Minas Gerais, Brazil, and to detect deoxyribonucleic acids (DNA) from bacteria of the genera Rickettsia and Borrelia through polymerase chain reaction (PCR) and 2) Detect molecular evidence of the presence of bacteria and protozoa of the genera Rickettsia, Borrelia , Babesia, Anaplasma and Ehrlichia in ticks collected from wild birds in the state of Rio de Janeiro. In the first study, 17 UC's were visited (national, state and municipal) where free-living ticks were captured using CO2 trap and manual collection. A total of 1,356 ticks were collected, including 1201 larvae, 106 nymphs and 49 adults. The nymphs were identified as Amblyomma spp. and adults such as A. oblongoguttatum, A. sculptum, A. brasiliense, A. dubitatum, A. aureolatum. The samples were submitted to DNA extraction by the phenolchloroform method with precipitation in isopropyl alcohol. In PCR using a partial sequence of the gltA gene as a target, Rickettsia spp. in 10.1% (19/188) of the samples, for Borrelia spp. a partial sequence of the flaB gene was used as a target and no sample was positive. Sequencing of 19 samples identified the presence of Rickettsia bellii DNA. For Chapter II, 33 birds of 20 different species were captured, of which 14 were parasitized by tick nymphs that were identified as A. longirostre. The presence of Rickettsia spp. in three samples, one of which was also positive for Borrelia spp. After sequencing, the co-infected sample showed 100% identity with Rickettsia bellii strain H3 and 91% with Borrelia turcica, two samples had 100% identity with Rickettsia sp. Aranha strain and AL strain. |

Aranha strain and AL strain.Este trabalho est? divido em dois cap?tulos e tem por objetivos: 1) Identificar as esp?cies de carrapatos na fase de vida livre, que ocorrem nas Unidades de Conserva??o (UC?s) dos estados do Rio de Janeiro (RJ), Esp?rito Santo (ES) e Minas Gerais (MG), e realizar detec??o de ?cidos desoxirribonucle?cos (DNA) de bact?rias dos g?neros Rickettsia e Borrelia por meio da rea??o em cadeia pela polimerase (PCR) e 2) Detectar evid?ncias moleculares da presen?a de bact?rias e protozo?rios dos g?neros Rickettsia, Borrelia, Babesia, Anaplasma e Ehrlichia em carrapatos coletados de aves silvestres no estado do Rio de Janeiro. No primeiro trabalho, foram visitadas 17 UC?s (nacionais, estaduais e municipais) onde carrapatos em fase de vida livre foram capturados utilizando armadilha de CO2 e coleta manual. Foram coletados 1.356 carrapatos, sendo 1201 larvas, 106 ninfas e 49 adultos. As ninfas foram identificadas como Amblyomma spp. e os adultos como A. oblongoguttatum, A. sculptum, A. brasiliense, A. dubitatum, A. aureolatum. As amostras foram submetidas a extra??o de DNA pelo m?todo de fenolclorof?rmio com precipita??o em ?lcool isoprop?lico. Na PCR utilizando como alvo uma sequ?ncia parcial do gene gltA, foi detectado DNA de Rickettsia spp. em 10,1% (19/188) das amostras, para Borrelia spp. foi utilizado como alvo uma sequ?ncia parcial do gene flaB e nenhuma amostra foi positiva. O sequenciamento das 19 amostras positivas na PCR, permitiu a identifica??o de DNA de Rickettsia bellii (KX0094101) em todas as amostras. Para o cap?tulo II, foram capturadas 33 aves, de 20 esp?cies diferentes das quais 14 estavam parasitadas por ninfas de carrapatos que foram identificados como A. longirostre. Foi detectado a presen?a de DNA de Rickettsia spp. em tr?s amostras, sendo uma delas positiva tamb?m para Borrelia spp.. This work is divided into two chapters and aims to: 1) Identify the species of ticks in the freeliving phase, which occur in the Conservation Units (UC's) in the states of Rio de Janeiro, Esp?rito Santo (ES) and Minas Gerais, Brazil, and to detect deoxyribonucleic acids (DNA) from bacteria of the genera Rickettsia and Borrelia through polymerase chain reaction (PCR) and 2) Detect molecular evidence of the presence of bacteria and protozoa of the genera Rickettsia, Borrelia , Babesia, Anaplasma and Ehrlichia in ticks collected from wild birds in the state of Rio de Janeiro. In the first study, 17 UC's were visited (national, state and municipal) where free-living ticks were captured using CO2 trap and manual collection. A total of 1,356 ticks were collected, including 1201 larvae, 106 nymphs and 49 adults. The nymphs were identified as Amblyomma spp. and adults such as A. oblongoguttatum, A. sculptum, A. brasiliense, A. dubitatum, A. aureolatum. The samples were submitted to DNA extraction by the phenolchloroform method with precipitation in isopropyl alcohol. In PCR using a partial sequence of the gltA gene as a target, Rickettsia spp. in 10.1% (19/188) of the samples, for Borrelia spp. a partial sequence of the flaB gene was used as a target and no sample was positive. Sequencing of 19 samples identified the presence of Rickettsia bellii DNA. For Chapter II, 33 birds of 20 different species were captured, of which 14 were parasitized by tick nymphs that were identified as A. longirostre. The presence of Rickettsia spp. in three samples, one of which was also positive for Borrelia spp. After sequencing, the co-infected sample showed 100% identity with Rickettsia bellii strain H3 and 91% with Borrelia turcica, two samples had 100% identity with Rickettsia sp. Aranha strain and AL strain. |
