Resumo: Given the relationship between various components of a formulation can positively or negatively interfere with the action of the active ingredient, the present study aimed to evaluate the influence of the volume of pour-on formulation containing fipronil used in dogs, maintaining the recommended dose of active ingredient, on the effectiveness against Ctenocephalides felis felis and Rhipicephalus sanguineus. For this purpose, 24 dogs were divided into four groups: control, which remained untreated, and treated groups A, B and C, medicated with 1.34 mL, 2.68 mL and 5.36 mL of a topical solution containing 10.7%, 5.3% and 1.8% fipronil, respectively. Each dog was infested two days before treatment and weekly after that with 100 samples of non-fed adult C. f. felis and 50 non-fed adults of R. sanguineus, keeping for both, equal sex ratio. The evaluation of immediate efficacy occurred two days after treatment and residual efficacy assessments were performed weekly, always 48 hours after each infestation. To this purpose, was proceeded the removal and counting of fleas and ticks, by manual grooming and/or by comb- test method, and the calculation of the efficacy made by Abbott?s formula. The group treated with 5.36 mL of 1.8% fipronil solution showed efficacy against R. sanguineus greater than 90% for 28 days, higher than groups treated with 1.34 mL and 2.68 mL of 10,7% and 5,3% fipronil solutions, which presented protection against re-infestation for 21 days period. In the two subsequent weekly assessments the highest volume remained superior to the others, although not reaching satisfactory levels of protection. Residual efficacy above 95% against C. f. felis was obtained up to 35 days by the three tested volumes. Despite fleacide efficacy less than 95% obtained in the 42? day after the treatment by all tested volumes, the 2.68 mL and 5.36 mL volumes were superior than 1.34 ml volume. It is concluded that the increased volume of topically solution of fipronil applied in dogs, maintaining the recommended dose of the active ingredient, increased its residual efficacy against C. f. felis and R. sanguineus, showing that simple changes in ectoparasiticide topical formulations may significantly modify the degree of protection provided

Para isso, foram utilizados 24 c?es divididos em quatro grupos: controle, o qual permaneceu sem tratamento, e os grupos tratados A, B e C, medicados com 1,34mL, 2,68mL e 5,36mL de solu??o contendo 10,7%, 5,3% e 1,8% de fipronil, respectivamente. Cada c?o foi infestado dois dias antes do tratamento e semanalmente ap?s o mesmo com 100 exemplares adultos n?o alimentados de C. f. felis e 50 adultos n?o alimentados de R. sanguineus, mantendo-se, para ambos, igual propor??o entre os sexos. A avalia??o de efic?cia imediata ocorreu dois ap?s o tratamento e as avalia??es de efic?cia residual foram realizadas semanalmente, sempre 48 horas ap?s as infesta??es. Para tal, procedeu-se a retirada e contagem das pulgas e carrapatos, por cata??o manual e/ou m?todo ?comb-test? e os c?lculos de efic?cia efetuados pela da f?rmula de Abbott. O grupo tratado com 5,36mL de solu??o de fipronil apresentou efic?cia contra R. sanguineus superior a 90% por 28 dias, sendo superior aos grupos medicados com 1,34mL e 2,68mL, os quais apresentaram-se protegidos contra reinfesta??es por um per?odo de 21 dias. Nas duas avalia??es semanais subsequentes o maior volume manteve-se superior aos demais, mesmo n?o atingindo n?veis satisfat?rios de prote??o. Efic?cia residual acima de 95% contra C. f. felis foi obtida por at? 35 dias pelos tr?s volumes testados. Apesar das efic?cias pulguicidas inferiores a 95% obtidas no quadrag?simo segundo dia ap?s o tratamento, os volumes de 2,68mL e 5,36mL apresentaram-se superiores ao volume de 1,34mL. description Given the relationship between various components of a formulation can positively or negatively interfere with the action of the active ingredient, the present study aimed to evaluate the influence of the volume of pour-on formulation containing fipronil used in dogs, maintaining the recommended dose of active ingredient, on the effectiveness against Ctenocephalides felis felis and Rhipicephalus sanguineus. For this purpose, 24 dogs were divided into four groups: control, which remained untreated, and treated groups A, B and C, medicated with 1.34 mL, 2.68 mL and 5.36 mL of a topical solution containing 10.7%, 5.3% and 1.8% fipronil, respectively. Each dog was infested two days before treatment and weekly after that with 100 samples of non-fed adult C. f. felis and 50 non-fed adults of R. sanguineus, keeping for both, equal sex ratio. The evaluation of immediate efficacy occurred two days after treatment and residual efficacy assessments were performed weekly, always 48 hours after each infestation. To this purpose, was proceeded the removal and counting of fleas and ticks, by manual grooming and/or by comb- test method, and the calculation of the efficacy made by Abbott?s formula. The group treated with 5.36 mL of 1.8% fipronil solution showed efficacy against R. sanguineus greater than 90% for 28 days, higher than groups treated with 1.34 mL and 2.68 mL of 10,7% and 5,3% fipronil solutions, which presented protection against re-infestation for 21 days period. In the two subsequent weekly assessments the highest volume remained superior to the others, although not reaching satisfactory levels of protection. Residual efficacy above 95% against C. f. felis was obtained up to 35 days by the three tested volumes. Despite fleacide efficacy less than 95% obtained in the 42? day after the treatment by all tested volumes, the 2.68 mL and 5.36 mL volumes were superior than 1.34 ml volume. It is concluded that the increased volume of topically solution of fipronil applied in dogs, maintaining the recommended dose of the active ingredient, increased its residual efficacy against C. f. felis and R. sanguineus, showing that simple changes in ectoparasiticide topical formulations may significantly modify the degree of protection provided
