Resumo: | Organochalcogens are important intermediates and the main applied reagents in organic synthesis, which can increase human exposure risk to these chemicals in the workplace. Since, the erythrocytic δ-ALA-D activity could be an important indicator of toxicity this report investigated the organochalcogens effects on blood δ-ALA-D in vitro and determined δ-ALAD activity on human occupationally exposed to organochalcogens. The acute tixicity of organochalcogens on erythrocytic δ-ALA-D activity, in mice, was also investigated. δ-ALA-D activity was assayed according to the method of Berlin & Schaller (1974) with some modifications. Diphenyl ditelluride, diphenyl diselenide and Ebselen inhibited in a concentration-dependent manner δ-ALA-D activity from human erythrocytes. Dithiothreitol (DTT), the hydrophobic SH-reducing agent, was able to reactivate and to protect inhibited δ-ALA-D. The administration of diphenyl diselenide (200 μmol/kg) and diphenyl ditelluride (150 μmol/kg) in a toxic amount inhibited the erythrocytic δ-ALA-D activity on mice. Otherwise, the human occupationally exposed to organochalcogens did not increase selenium levels, from these findings we can not affirm that human δ-ALA-D was a target for organochalcogens ex vivo. In addition δ-ALA-D was a target toxicity from organochalcogens, in vitro. But, mice erytrocytes δ-ALA-D was a target for organochalcogens tested ex vivo. |

Devido ao fato de que a enzima δ- ALA-D pode ser um indicador de toxicidade a organocalcogênios, o presente trabalho visa avaliar o efeito do disseleneto de difenila, ditelureto de difenila e Ebselen sobre a atividade da enzima δ-ALA-D sanguínea in vitro e também determinar a atividade desta enzima em pessoas expostas ocupacionalmente a estes compostos. Investigou-se também o efeito da administração de doses tóxicas de organocalcogênios sobre a atividade da δ-ALA-D sanguínea de camundongos. A dosagem da atividade enzimática foi determinada de acordo com o método de Berlim & Schaller (1974), com algumas modificações. Os níveis sangüíneos de selênio e parâmetros hematológicos foram determinados. O disseleneto de difenila, ditelureto de difenila e Ebselen inibiram de maneira dependente da concentração a atividade da enzima da δ-ALA-D sanguínea de humanos, in vitro. O ditiotreitol, um agente redutor sulfidrílico, foi capaz tanto de proteger quanto de reverter a inibição enzimática causada pelos organocalcogênios, sugerindo que a inativação da δ-ALA-D sanguínea in vitro ocorre devido a interação com grupos sulfidrílicos essenciais da enzima. A administração de doses tóxicas de disseleneto de difenila (200 μmol/kg) e ditelureto de difenila (150 μmol/kg) inibiram a atividade da δ-ALA-D sanguínea de camundongos. Por outro lado, a atividade desta enzima apresentou um aumento de 30% no grupo de pessoas expostas ocupacionalmente a organocalcogênios. Uma vez que o grupo exposto ocupacionalmente aos organocalcogênios não mostrou níveis sanguíneos de selênio aumentados, não é possível afirmar que a δ-ALA-D é um alvo para os organocalcogênios ex vivo, devido a isso utilizou-se um modelo de intoxicação animal.
