Resumo: The sodium citrate is one of the most utilized in chemical food additives that have been found in tinned and milk derivatives. Its proprieties are: stabilizer, emulsifier and acidity regulator. The chemical compound can be dissociated in water by releasing its anion. The endogenous citrate in biological systems plays an important role in the general metabolism, serving as a source of acetyl-CoA that can be utilized for the synthesis of lipids and acetylcholine. The citrate also has an important role in controlling the effects on enzymes as Phosphofructokinase and Acetyl-CoA carboxylase. The aims of this study are the investigations on the effects of the additive over lipid and acetylcholine metabolism together with the evaluation of biochemical parameters with ions Na+, K+, urea and creatinine. The mice were separated in two different treatments: subchronic (55 days) and chronic (110 days), males and females. The sodium citrate solutions were control (potable water), 1%, 2.5% and 5% ad libitum in drinking water. The results showed an increase of the body weight in 2.5% concentration for both subchronic and chronic treatments. In the inner organs, the study showed a decrease of livers weight in 5% concentration. In the histological analysis we observed that the animal cells in 2.5% concentration had more capacity of lipid saving, but in 5% this capacity is decreased. It was observed an increase on the quantify of muscle lipids on the male in both treatments. The lipase evaluation has shown a decrease on the subchronic males in 2.5% and 5% sodium citrate concentrations. Didn’t appear changes in K+ levels. In the ACh metabolism had an increase of AChE in the subchronic male and chronic female in vivo treatments. The in vitro experiments had a tendency for activity increasing AChE in sodium citrate 50 mM. The objective of this work was to recover aspects around the application of sodium citrate and its possible interactions in lipid metabolism and acetylcholinesterase activity.

Suas propriedades são de estabilizante, emulsificante e regulador de acidez. O composto químico pode ser dissociado em meio aquoso liberando seu ânion. Já o citrato endógeno nos sistemas biológicos exerce um importante papel no metabolismo em geral, pois tem a capacidade de suprir o conteúdo citoplasmático de unidades de acetil-CoA que podem ser utilizadas para a produção de lipídios e da acetilcolina. O citrato também exerce efeitos importantes modulando enzimas como a fosfofrutoquinase e acetil-CoA-carboxilase. No presente estudo investigou-se o efeito do aditivo sobre o metabolismo lipídico e da acetilcolina juntamente com a avaliação de parâmetros bioquímicos como os íons Na+, K+, uréia e creatinina. Os camundongos foram divididos em tratamentos subcrônicos (55 dias) e crônicos (110 dias), machos e fêmeas. As soluções de citrato de sódio foram controle (água potável), 1%, 2,5% e 5% ad libitum na água de beber. Os resultados demonstraram um aumento dos pesos corporais na concentração 2,5%, e uma diminuição dos pesos em 5% em ambos os tratamentos subcrônico como crônico. Na pesagem dos órgãos internos, evidenciamos a diminuição dos pesos dos fígados em 5%. Frente à análise histológica observamos que as células dos animais 2,5% possuem maior capacidade de armazenamento lipídico, já em 5% a capacidade é diminuída. Na quantificação dos lipídios musculares, foi observado uma maior quantidade de lipídios nos machos de ambos os tratamentos 2,5%. A avaliação da lipase plasmática, demonstrou uma diminuição nos machos subcrônicos 2.5% e 5%. Também, houve uma diminuição dos triglicerídeos plasmáticos dos grupos em 5%. As análises bioquímicas demonstram um aumento no Na+ plasmático em 5% bem como um aumento na uréia e na creatinina nos grupos citrato de sódio 5%. Não houveram alterações na concentração plasmática do íon K+. Frente ao metabolismo da ACh percebe-se um aumento da AChE nos tratamentos machos subcrônicos e fêmeas crônicas in vivo. No experimento in vitro notamos uma tendência ao aumento da AChE na concentração final 50 mM de citrato de sódio.
