Resumo: The aim of this study was evaluating the testosterone suppression impact on craniofacial growth in rats. Thirty-six Wistar rats were included in this study. Orchiectomy (ORX) was performed in experimental group (n=8), while sham surgery was performed in control group (n=18) with 23 days old. The animals were allocated in 4 groups: an ORX group (n=9) and a sham group (n=9) euthanized with 45 days old and an ORX group (n=9) and a sham group (n=9) euthanized with 73 days old. Radiographic Lateral Cephalograms were performed on all animals with 23, 45 and 73 days old. Cephalometric measurements were obtained and compared with t-student and Mann-Whitney tests performed with GraphPad Prism software. The p<0,05 values were statistically different. After euthanasia of the groups, the mandibular condyle, the pre-maxillary suture and the middle palatal suture were processed for qualitative histological analysis of sagittal and frontal sections stained in Hematoxylin and Eosina and Picrosírius and Hematoxylin. The images were obtained using conventional light microscopy and polarized light. Body weight was lower in ORX groups. Verticals and horizontals cephalometric measurements were lower in ORX groups. Cephalometric angles were also statistically different in ORX groups. Histological analyzes demonstrate a reduction in the differentiation of chondroblast precursor cells in the mandibular condyle in orchiectomized animals, in addition to the lower proliferation of osteoblasts and formation of bone trabeculae. There is a higher concentration of collagen fibers in the Sham groups compared to the ORX groups in the mandibular condyle, and type III collagen fibers were observed only in the Sham groups. In the median palatine suture, greater proliferation of osteoblast precursor cells was observed in animals of the control group. Type III collagen fibers were also observed only in the Sham groups. There were no significant differences in the premaxillary suture. Concludes that the testosterone suppression decreases the craniofacial growth during puberty of rats

Dessa forma, o objetivo deste estudo foi avaliar o impacto da supressão de testosterona sobre o crescimento e desenvolvimento craniofacial de ratos. Trinta e seis ratos da linhagem Wistar foram selecionados para este estudo. Orquiectomia (ORX) foi realizada nos animais do grupo experimental (n=18) e cirurgiam sham nos animais do grupo controle (n=18) no 23º dia de vida. Os animais foram alocados em 4 grupos: um grupo ORX (n=9) e um grupo sham (n=9) eutanasiados aos 45 dias de vida, e o outro grupo ORX (n=9) e o outro sham (n=9) eutanasiados aos 73 dias de vida. Tomadas radiográficas laterais do crânio foram realizadas em todos os animais nos 23º, 45º e 73º dia de vida. Medidas cefalométricas e o peso corporal foram obtidos e comparados pelo teste t de Student ou Mann-Whitney pelo software GraphPad Prism. Valores de p<0,05 foram considerados significativos. Após eutanásia dos grupos, o côndilo mandibular, a sutura pré-maxilar e a sutura palatina mediana foram processadas para análise histológica qualitativa de cortes sagitais e frontais corados em Hematoxilina e Eosina e Picrosírius e Hematoxilina. As imagens foram obtidas em microscopia de luz convencional e luz polarizada. O peso corporal foi estatisticamente menor nos grupos ORX (p < 0,05). Medidas cefalométricas verticais e horizontais foram menores nos grupos ORX. O comprimento do osso nasal, a altura craniana anterior, a altura mandibular e as medidas cefalométricas entre os pontos 3-5 e 21-23 foram estatisticamente menores nos grupos ORX em ambos os tempos experimentais. Ângulos cefalométricos também foram estatisticamente diferentes nos grupos ORX. As análises histológicas demonstram uma redução da diferenciação de células precursoras de condroblastos no côndilo mandibular em animais orquiectomizados, além de menor proliferação de osteoblastos e formação de trabéculas ósseas. Há maior concentração de fibras colágenas nos grupos Sham em comparação aos grupos ORX no côndilo mandibular, e fibras colágenas do tipo III foram observadas apenas nos grupos Sham. Na sutura palatina mediana, foi observada maior proliferação de células precursoras de osteoblastos em animais do grupo controle. Fibras colágenas tipo III também foram observadas apenas nos grupos Sham. Não foi observado diferenças significativas na sutura pré-maxilar.
